Influence of Side Chain Conformation on the Activity of Glycosidase Inhibitors

Abstract: Substrate side chain conformation impacts reactivity during glycosylation and glycoside hydrolysis and is restricted by many glycosidases and glycosyltransferases during catalysis. We show that the side chains of gluco and manno iminosugars can be restricted to predominant conformations by strategic installation of a methyl group. Glycosidase inhibition studies reveal that iminosugars with the gauche, gauche side chain conformations are 6-to 10-fold more potent than isosteric compounds with the gauche, trans con… Show more

“…Both therapies in this study significantly reduced nitric oxide levels, suggesting they may mitigate the nitrative stress caused by peroxynitrite in the brain. The findings of this study support previous claims that combination medication may be more effective than monotherapy [ 94 , 95 , 96 , 97 ]. Treatment with jambolan extract containing gallic acid considerably improved the brain’s antioxidant status compared with treatment with aluminum chloride alone in the animals utilized in this investigation.…”

“…Modern medicine has grown in relation to efforts towards the development of antidiabetic medicine such as glycoside inhibitors as a way to reduce the absorption of carbohydrates so as to lower postprandial glucose and insulin levels [ 17 , 18 ].…”

“…One of the main targets for the search for new antidiabetic drugs is the inhibition of intestinal glucosidases. Iminosugars synthesized and sugar derivates are potential antidiabetic drugs, showing a favorable impact in glycosidase inhibition, improving their activity through inhibitor synthesis and design of conformational preorganization [ 8 , 9 ]. On the other hand, medicinal plants are an excellent alternative to finding complementary treatments to achieve improved control of hyperglycemia [ 10 ].…”
