Influence of polygenic risk scores for schizophrenia and resilience on the cognition of individuals at-risk for psychosis

Abstract: Cognitive impairment is a core feature of schizophrenia which precedes the onset of full psychotic symptoms, even in the ultra-high-risk stage (UHR). Polygenic risk scores (PRS) can be computed for many psychiatric disorders and phenotyping traits, including scores for resilience. We explored the correlations between several PRS and neurocognition in UHR individuals. We included 107 UHR individuals; 29.9% of them converted to psychosis (UHR-C) while 57.0% did not (UHR-NC) during the 1-year follow-up. Cognitive… Show more

“…The literature shows that the GPSs of cognitive performance and educational attainment are significantly associated with general intelligence 14, 15 and brain development 14, 16 . In addition, the GPSs of schizophrenia and ADHD successfully predicted psychiatric disorders 1719 . However, the combined causal effects of genes and the neighborhood environment during development in children remain unclear.…”

“…A systematic review by Ribeiro-Santos, Teixeira and Vinicius [ 45 ] found that MCP-1 and IL-18 levels were associated with cognitive impairments in schizophrenia. In addition, Lim et al [ 32 ], Perkins et al [ 46 ], and He et al [ 47 ] reported PRS as a potential biomarker for early cognitive deficits or for the transition from the ultra-high-risk state to established psychosis.…”

“…Self-efficacy 16 is an overall relatively stable sense of competence or self-confidence that individuals show in coping with challenges in various environments. Zhou 46 indicated that self-efficacy and motivation may act as mediators in the relationship between neurocognition and self-management in schizophrenia.…”

“…14 Abroad, Gooding PA 15 proposed improving resilience, which could allow patients to resist negative ideas and impulses, such as self-injury or suicide, and highlighted external factors that supported resilience, including social support, reciprocity and religious coping. He 16 suggested that cognitive performance and a high level of resilience are positively correlated and that improving resilience can improve cognitive function. Cognitive impairment is a core feature of schizophrenia.…”
