River Nile represents the most important source of freshwater in Egypt. There are several factors lead to the water pollution in the River Nile System (main stream Nile, drains and canals). The water quality of River Nile was good despite high organic loads discharged from some of the drains and industrial activities. These were harmful both to human and stream ecosystem health. A biological approach to monitoring water quality incorporates use of stream organisms themselves as a basis for pollution detection. Fish and aquatic insects considered as bio-indicators of pollutant effects and help to investigate possible environmental problems. One of the recent biochemical techniques for detecting DNA damage as a result of DNA single strand breaks, alkali-labile sites, and cross-linking is the alkaline (pH>13) single cell gel SCG assay. In this study the comet assay measure the DNA damage in various stages of the mosquito Culex pipiens collected from two different polluted water streams (Nikla and Elmansoreyh). The DNA damage measured in 3 rd , 4 th larval instars, pupa, male and female adults. It is clear from the results obtained in this study that the genotoxicity of water pollution of two different polluted water streams (Nikla and Elmansoreyh) in C. pipiens was high in larval and pupal stages and this study affirmed the appropriateness of the comet assay as a sensitive tool for environmental monitoring. Additionally, it can be proposed that C. pipiens is a strong aquatic bioindicator of genotoxicity.

Long lasting exposure of animals to stressing factor may lead to the selection of population able to cope with the stressor at lower cost than unexposed individuals. The aim of this study was to assess whether 130-generational selection of a beet armyworm to cadmium in food might have induced tolerance also to other stressors. The potential tolerance was assessed by means of unspecific stress markers: HSP70 concentration, DNA damage level, and energy budget indices in L5 larval instars of beet armyworm. The animals originated from Cd-exposed and control strains exposed additionally in a short-term experiment to high/low temperature or pesticide-spinosad. The application of the additional stressors caused, in general, an increase in the levels of studied parameters, in a straindependent manner. The most significant increase was found in HSP70 level in the individuals from the Cd-strain exposed to various spinosad concentration. Therefore, multigenerational contact with cadmium caused several changes that enable the insect to survive under a chronic stress, preparing the organism to the contact with an additional, new stressor. This relationship may be described as a sort of cross tolerance. This may, possibly, increase the probability of population survivorship and, at the same time, decrease the efficiency of pesticide-based plant protection efforts.

Purpose: To analyze the role of mus-genes repair in system activation P-elements in Drosophila melanogaster induced by a chronic exposure to low doses. Materials and methods: The materials were dysgenic individuals of Drosophila melanogaster with mutations in repair genes (mus101, mus205, mus304, mus308, mus309) and simultaneous transposition of mobile P-elements. The animals were exposed to a chronic irradiation in low doses (0.42 mGy/h). The reaction of animals was analyzed by the DNA damage rate in somatic cells ('Comet assay'), level of dominant lethal mutations, fecundity, and survival rate. Results: The combined action of the systems of post-replication, recombination repair (mus205, mus304), repair of DNA double-stranded breaks (mus304), and of the transposition activity of P-elements after a chronic irradiation in low doses was identified according to every study parameter. The other repair systems and their genes (mus101, mus308, and mus309) responded to action of only one factor (irradiation or mobile elements transposition). Conclusion: The obtained data significantly contribute to the knowledge on a new reaction of the mechanisms of organisms to a chronic irradiation in low doses. ARTICLE HISTORY
