Interaction between gene repair and mobile elements-induced activity systems after low-dose irradiation

Abstract: Purpose: To analyze the role of mus-genes repair in system activation P-elements in Drosophila melanogaster induced by a chronic exposure to low doses. Materials and methods: The materials were dysgenic individuals of Drosophila melanogaster with mutations in repair genes (mus101, mus205, mus304, mus308, mus309) and simultaneous transposition of mobile P-elements. The animals were exposed to a chronic irradiation in low doses (0.42 mGy/h). The reaction of animals was analyzed by the DNA damage rate in somatic … Show more

“…The biological efficiency of low dose irradiation could be higher than the exposure to acute irradiation (Grosovsky, 1999;Ma et al, 2010;Yushkova and Zainullin, 2016). The main effect of low doses is radiation-induced genetic instability which lasts during cell divisions after the cell has been irradiated (Holmberg et al, 1998;McIlrath et al, 2003).…”

“…This is confirmed not only by numerous studies of other authors, but by our own experimental facts. Both irradiation and mutations in stress-response genes have been shown to affect the effects of TE transpositions under dysgenic conditions (Yushkova and Zainullin, 2016). This is reflected in an increase of sterility, the frequency of locus-specific mutability, recessive and dominant lethal mutations (Margulies et al, 1986(Margulies et al, , 1987Yushkova and Zainullin, 2014).…”

“…DNA damage in neuroblasts and midgut cells of F 1 /F 2 third instar larvae was assessed using the Comet assay (Yushkova and Zainullin, 2016). The cell suspension was obtained by treating somatic tissues to a collagenase solution (type IV, 0.5 mg/ml PBS) at 37 • C. The obtained cells of the nerve ganglia and midgut (10 µl) were mixed with 0.5% low melting point agarose (100 µl) and applied on glass slides coated with a layer of 1% normal melting point agarose.…”
