An 8‐week growth trial was conducted to evaluate effects of dietary oil sources on growth, enzymes activity and genes expression levels related to lipid metabolism of hybrid grouper (♀Epinephelus fuscoguttatus × ♂E. lanceolatu) juveniles. Seven iso‐lipid (97 g/kg of dry matter) and iso‐protein (503.5 g/kg of dry matter) experimental diets were formulated containing 50 g/kg fish oil (FO; acting as controls) or various vegetable oils (VOs): corn oil (CO), sunflower oil (SO), tea oil (TO), olive oil (OO), rice oil (RO) and mixed oil (MO; comprising equal amounts of these oils). Each diet was fed to triplicate groups of 40 fish for per repetition (15.09 ± 0.01 g) for 56 days. The results show that (a) alternative dietary oils had no significant effects on final weight compared with control group (p > .05); (b) compared with FO group, VOs significantly changed the contents of serum lipoproteins, cholesterol, triglycerides and the activity of liver lipid‐metabolizing enzymes (p < .05); (c) CO group had the least effect on the serum lipoproteins, triglycerides and cholesterol of grouper compared with control; the activity of liver lipid‐metabolizing enzymes in RO and control group was the closest; (d) the mRNA levels of Δ6 Fatty acid desaturase (Δ6Fad), hormone‐sensitive lipase (HSL) and lipoprotein lipase (LPL) were not significantly effected by lipid sources, but CO, TO, OO and MO significantly down‐regulated the expression of fatty acid synthetase (FAS) mRNA level in liver, while RO opposite (p < .05); (e) vegetable oil significantly up‐regulated peroxisome proliferator‐activated receptor α (PPARα) and peroxisome proliferator‐activated receptor β (PPARβ) mRNA levels, while TO and RO down‐regulated peroxisome proliferator‐activated receptor γ (PPARγ) mRNA levels (p < .05); and 6) MO significantly increased the mRNA levels of heart‐type fatty acid‐binding protein (H‐FABP) and adipocyte‐type fatty acid‐binding protein (A‐FABP) (p < .05), while other VOs had no effect on them (p > .05). In conclusion, dietary substitution of FO by VO in diet affected lipid metabolism of grouper, which may be regulated by PPARs.

The aim of this study was to investigate the effects of substituting soybean oil (SO) for fish oil (FO) on the performance, tissue fatty acid (FA) composition, plasma components, liver metabolic enzyme activity and mRNA levels of genes related to lipid metabolism in the liver of groupers (Epinephelus coioides). We formulated five isolipidic and isoproteic diets with increasing SO levels (0, 250, 500, 750 and 1000 g/kg, respectively). Triplicate groups of 30 groupers (initial mean body weight of 12.6 g/ fish) were fed one of the diets twice daily, to apparent satiety across a feeding period of 56 days. The growth performance, whole-body composition, and protein and lipid muscle contents did not differ across the dietary treatments. In contrast, the liver lipid content had positive linear and quadratic responses to the increasing dietary SO levels, but the liver protein content had the opposite trend, and the highest lipid value and lowest protein value occurred in the 250 and 1000 g/kg SO diets, respectively. There were no significant effects of increasing dietary SO inclusion levels on any of the plasma components and parameters of liver metabolic enzyme activity, except for acetyl coenzyme A carboxylase and hepatic lipase, which showed linear and quadratic responses to increasing dietary SO inclusion levels and peaked at 1000 and 250 g/ kg SO inclusion level, respectively. However, the FA profiles of the dorsal muscle and liver generally reflected the FA profile of the diet. Furthermore, the mRNA levels of fas, acc, g6pd, lpl, pparα, cpt-1, srebp-1c, δ6fad and elovl5 in the liver exhibited positive linear and/or quadratic responses to dietary SO inclusion levels. However, negative linear and/or quadratic responses were observed for the mRNA levels of hsl and atgl in the liver, with increasing dietary SO inclusion levels. The mRNA levels of scd and fabp in the liver were not affected by the dietary SO inclusion level. These results indicate that FO could be replaced completely by SO without affecting growth. However, the inclusion of SO at levels higher than 500 g/kg could compromise the FA profile in the liver and flesh of the fish species. To study the effects of acute ammonia stress on the poisoning reaction of the hybrid grouper (♀ Epinephelus fuscoguttatus × ♂ E. lanceolatu), 300 healthy grouper juveniles with an initial body weight of 51.4 ± 2.57 g were selected for an acute ammonia stress experiment using a half-lethal concentration of ammonia for 24, 48, 72, and 96 h with triplicate. The results show that (1) The half-lethal concentrations of ammonia for a hybrid grouper were 39.5 mg/L for 24 h, 27.3 mg/L for 48 h, 26.5 mg/L for 72 h, and 25.0 mg/L for 96 h, and the safe concentration was 2.50 mg/L. (2) The biochemical indices of the serum and livers of the groupers fluctuated under acute ammonia stress (P < 0.05), and the contents of serum cortisol and lactate reached the maximum value in 72 h (P < 0.05). (3) 24 h ammonia stress increased the activities of alkaline phosphatase, acid phosphatase, catalase, glutathione peroxidase, superoxide dismutase, lysozyme, and the content of immunoglobulin M in the liver (P < 0.05). Meanwhile, all ammonia-stressed fish groups had an increased amount of malondialdehyde in the liver (P < 0.05). (4) The ammonia poisoning caused significantly up-regulation of antioxidant genes, inflammatory cytokines, and apoptosis genes (P < 0.05), and the expression of inflammatory cytokines and apoptosis genes were the highest in 24 h ammonia stress group. (5) The ammonia content in the water changed the abundance and evenness of intestinal microbes in grouper, mainly in that ammonia stress significantly increased the relative abundance of fusobacteria, but significantly decreased the relative abundance of gemmatimonadetes (P < 0.05). It was speculated that apoptosis induced by ammonia poisoning in grouper may be related to oxidative stress and the activation of inflammatory factors, and excessive inflammatory stress may be one of the causes of ammonia poisoning in the hybrid grouper.
