Industrial application of QbD and NIR chemometric models in quality improvement of immediate release tablets

Abstract: Quality by Design (QbD) and chemometric models are different sides of the same coin. While QbD models utilize experimentally designed settings for optimization of some quality attributes, these settings can also be utilized for chemometric prediction of the same attributes. We aimed to synchronize optimization of comparative dissolution results of carvedilol immediate release tablets with chemometric prediction of dissolution profile and content uniformity of the product. As an industrial application, selectio… Show more

“…This implies that the constructed models can account for 70%, 60%, and 60% of the variation in the responses of C2H2, C3H6, and C6H6, respectively. In other words, changing the measurement location cannot explain much of the variance of the calculated ion currents of these gases [50]. Further confirmation of the good model fitness was checked through the CV% (coefficient of variation).…”

“…The key to implementing QbD is an understanding of the product process, which requires modeling the process. 8 The purpose of modeling is to investigate the impact of various variables in the formulation process on product design indicators such as crucial material attributes (CMAs) and crucial process parameters (CPPs). 9 Producing final dosage forms, usually in the form of oral solid dose (OSD) medications such as tablets, requires a series of complex unit operations.…”

“…This term is also known the Design of Experiments (DoE). [1,16,17] QbD promotes pharmaceutical organizations want to put into effect quality risk control concepts to benefit a higher understanding in their production strategies and make sure higher product quality. [2] Carbimazole is an anti-hyperthyroid drug commonly used to treat Graves' disease, subacute thyroiditis due to toxic nodular goiter, and radioactive iodine therapy.…”
