Induction of tumor necrosis factor and interleukin-1 by purified staphylococcal toxic shock syndrome toxin 1 requires the presence of both monocytes and T lymphocytes

Abstract: Highly purified staphylococcal toxic shock syndrome toxin 1 (TSST-1) was tested for its ability to induce the cytokines tumor necrosis factor (TNF) and interleukin-1 (IL-1) from fractionated human peripheral blood mononuclear cells prepared from seven healthy donors. Highly purified monocytes alone or T lymphocytes alone did not produce TNF or IL-1 when incubated with TSST-1 at 37°C for up to 72 h. However, the addition of 10 lag of TSST-1 per ml to a 1:1 mixture of monocytes and T cells resulted in significan… Show more

“…Thus, both cell types are required for an efficient response. A similar requirement for the presence of both monocytes and lymphocytes together has been reported elsewhere for induction of TNF-␣ by SEA (15) and TSST-1 (42). Moreover, incubation of monocytic cell line THP-1 with SEB did not activate TNF-␣ gene expression (31).…”

“…TNF-␣ induced by superantigen can be produced by both monocytes and T cells (1,15,30). Previous studies have examined the induction of TNF-␣ by SEA, SEB, or TSST-1 (15,30,38,42,43). In this study, we wished to characterize the induction of TNF-␣ by SEB in mixed cultures of human monocytes in the presence of lymphocytes.…”

“…Monokine expression presumably requires participation of bofh T cells and antigen-presenting cells (APC) (See et al, 1992). Purified monocytes stimulated with several superantigens produce little TNF-a or tl-ip in fhe absence of viable T cells or y-interfercn (Kotb, 1993).…”

“…Like other staphylococcal toxins TSST-1 has numerous immunomodulating effects such as inducing the release of interleukin 1 and the tumour necrosis factor from monocytes (Yokomizo et al 1995). Its mitogen effect on T-lymphocytes results in the release of lymphokines (See et al 1992). When bovine T-lymphocytes are experimentally exposed, TSST-1 acts as a superantigen for bovine immune cells, thus it potentially contributes to the mammary pathology associated with Staph.…”
