Changes in oxytocin (OT) receptor expression have been found to be an important determinant of the physiological effect of OT in several species. To date there are no published studies of OT binding sites during pregnancy in the pig. The purpose of the present study is to improve understanding of the role of OT in porcine parturition. The concentration and affinity of OT binding sites were determined for myometrium and endometrium from pregnant and postpartum gilts. Tissues were obtained after slaughter from 7 animals in each of four groups: 1) 90 days gestation, 2) 112 days gestation, 3) term after milk letdown (before delivery), and 4) within 1-3 h after farrowing. Mammary tissues were obtained for some animals in each group (n = 3-5/group). Before slaughter, blood was collected from each animal and assayed for estradiol-17 beta, progesterone, 13,14-dihydro-15-keto-prostaglandin F2 alpha (PGFM), and OT. Binding of 3H-OT in the three tissues was concentration- and time-dependent. Sites of 3H-OT binding (fmol/mg protein +/- SEM) increased toward term for each tissue and remained elevated in the postpartum group. Endometrial and mammary tissues displayed the most acute increases in OT binding site concentrations while myometrial tissues displayed a more gradual increase in OT binding sites over the times studied. The binding sites displayed high affinity for 3H-OT and were characterized by linear Scatchard plots. Concentrations of estradiol-17 beta, PGFM, and OT (pg/ml +/- SEM) were positively correlated with 3H-OT binding site concentrations, whereas progestrone concentrations (ng/ml +/- SEM) were negatively correlated with binding site concentration, as determined by Pearson's Correlation Analysis. The data represent the first account of changes in the expression of OT binding sites on porcine tissues during gestation and labor.

Partus induction is an important integrative component of reproduction management in the swine industry. In this context, the question is to what extent the application of uterotonic substances may influence the vitality of piglets. The blood pH value is used as a laboratory objective diagnostic parameter of neonatal acidosis. This parameter of the acid-base balance was examined in piglets born spontaneously at the 115th to 116th day of gestation (group 1, n = 57) and after two different partus induction protocols starting at the 114th day of gestation (group 2: combination of 175 microg of cloprostenol and 20 IU of oxytocin, n = 55; group 3: 175 microg of cloprostenol and 35 microg of carbetocin, n = 56). Initial blood samples were taken from each piglet at birth and 2 h later. The results show that the different protocols have no negative influence on the initial pH value and the compensation of neonatal acidosis until the second hour post-natum.

This paper will examine the augmentation of oxytocin (OT) secretion in the final day before parturition in the pig, and review the key endocrine systems of influence. Recent experiments have shown that increases in plasma OT above baseline secretion were not seen until 7 hours prior to the birth of the first piglet. Progesterone release begins to decline some days before birth and is unlikely to provide such acute control of OT release immediately before birth. The involvement of relaxin is complex but it is possible that the prepartum relaxin surge, which ends close to the OT rise, attenuates OT secretion to prevent excessive myometrial activity until cervical softening has occurred. Increasing myometrial electromyographic activity appears to be seen as relaxin concentrations decline and OT increase. Endogenous opioid peptides within the hypothalamus exert a profound influence upon OT secretion during birth, and some effects are also seen prepartum. It has been proposed that this system may allow parturition to be interrupted or slowed in response to changes in environment. The therapeutic choice of exogenous injections of OT to control the onset time of parturition will be discussed in the light of current physiological knowledge.
