Induction of Nitric Oxide Synthase Activity in Adrenal Cells

Abstract: The induction of nitric oxide synthase (NOS) II by bacterial lypopolysaccharide (LPS) was studied in a steroidogenic mouse tumor adrenal cell line (Y1). Conditioned media from LPS-stimulated peritoneal macrophages induced an increase in NOS II expression as shown by western and northern blot analysis. Accordingly, in the presence of conditioned media an increase in nitrite levels was observed. In addition, steroid production was significantly decreased. In conclusion, NOS II expression could be induced in ster… Show more

“…Previous studies from our laboratory demonstrated the expression of both constitutive NOS isoforms and the induction of NOS-2 by lipopolysaccharide (LPS) in adrenal cells (Cymeryng et al 2000(Cymeryng et al , 2002. We have also presented evidence supporting a modulatory role for NO on adrenal steroidogenesis: NO donors (Cymeryng et al 1998) and L-arginine (Cymeryng et al 1999) significantly inhibit steroid synthesis by a mechanism that probably involves a direct interaction of NO with CYP11A1 (cytochrome P450scc).…”

“…Steroid-producing cells within the adrenal cortex constitutively express NOS-1, NOS-3, and HO-2 enzymes, and respond to an immune challenge by increasing NOS-2 expression and activity (Cymeryng et al 2000, Pomeraniec et al 2004. As discussed in previous reports from our laboratory (Cymeryng et al 2002), the obtained ZF-R fragments necessarily contain different proportions of endothelial, neuronal, and immune cells displaying NOS and/or HO activity.…”

“…22 NO is produced endogenously from L-arginine by the NO synthases (NOS) enzyme family, mainly in vascular endothelial cells. 23,24 Thyroid hormones (THs) influence endothelium and directly rise NO production in vascular smooth muscle cells through the phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signallingpathway. 25 Direct measurement of this ubiquitous free radicalsignalling molecule is impossible due to its short half-life (< 0.1 s).…”

“…Owing to the use of nitrate as an agricultural fertilizer and another atmospheric release of nitrogen oxides, environmental nitrogen emissions have increased enormously (3), leading to an increase in the nitrogen concentrations of surface waters as a potential water pollutant (1). NO is produced endogenously from L-arginine by NO synthases (NOS) enzyme family, mainly in vascular endothelial cells (4,5). Thyroid hormones (THs) influence endothelium and directly rise NO production in vascular smooth muscle cells through the phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway (5,6).…”
