Induction Of Dna Synthesis In Lymphocytes In Vitro By Various Bacteria, With Special Reference To Neisseria Gonorrhoeae, In Patients With Uro-Arthritis (Reiter's Disease)

Abstract: Antigen-induced lymphocyte proliferation was studied by quantitation of 14C-thymidine uptake in cell cultures. The induction of DNA synthesis in vitro in lymphocytes from patients with uro-arthritis after stimulation by whole cells of virulent and avirulent N. gonorrhoeae, meningococci group B, and apathogenic Neisseria (N. pharyngis) was compared with the DNA synthesis in lymphocytes from healthy controls after stimulation with the same Neisseria antigens. The difference between patients and the controls was … Show more

“…Pathologic specimens suggested that events similar to those noted at mucosal surfaces occurred after dissemination. For example, gonococci have been seen inside A cells of the synovial membrane (14), and gonococcal peptidoglycan monomers have been implicated in cell damage at synovial membranes (42,47 …”

“…Gonococci have many antigens in common with meningococci. Rosenthal and Danielsson (1978) found a close antigenic relationship between these microorganisms. Therefore the possibility of an immune response developing as a result of meningococcal infection cannot be excluded.…”

“…The bacterial antigens were prepared by Dan Danielsson, Orebro, Sweden. The procedures used have been described in detail elsewhere (Rosenthal and Danielsson, 1978). PPD tuberculin was obtained from Statens Seruminstitut, Copenhagen, as a solution of 1 mg/ml in phosphate buffer, without addition of preservative.…”

“…A dilution of 1:500 of this antigen was found to be optimal in pilot tests. Later on, one strain of Neisseria gonorrhoeae of colony morphology type T2 whole cells was prepared as described elsewhere (Rosenthal and Danielsson, 1978). at a concentration of approximately 2 x 106 per culture.…”

“…Triplicate cell cultures containing 3 x 105 E cells/tube with and without antigen were set up. E 0-1 ,uCi of '4C-thymidine in 0 05 ml of phosphate buffered saline was added to each culture on day 5, and the radioactivity was measured in an Intertechnique (Nano teknik, Sweden) liquid scintillation X~c ounter (Rosenthal and Danielsson, 1978).…”
