We provide a transcriptional profile of coffee rust interaction and identified putative up regulated resistant genes Coffee rust disease, caused by the fungus Hemileia vastatrix, is one of the major diseases in coffee throughout the world. The use of resistant cultivars is considered to be the most effective control strategy for this disease. To identify candidate genes related to different mechanism defense in coffee, we present a time-course comparative gene expression profile of Caturra (susceptible) and Híbrido de Timor (HdT, resistant) in response to H. vastatrix race XXXIII infection. The main objectives were to obtain a global overview of transcriptome in both interaction, compatible and incompatible, and, specially, analyze up-regulated HdT specific genes with inducible resistant and defense signaling pathways. Using both Coffea canephora as a reference genome and de novo assembly, we obtained 43,159 transcripts. At early infection events (12 and 24 h after infection), HdT responded to the attack of H. vastatrix with a larger number of up-regulated genes than Caturra, which was related to prehaustorial resistance. The genes found in HdT at early hours were involved in receptor-like kinases, response ion fluxes, production of reactive oxygen species, protein phosphorylation, ethylene biosynthesis and callose deposition. We selected 13 up-regulated HdT-exclusive genes to validate by real-time qPCR, which most of them confirmed their higher expression in HdT than in Caturra at early stage of infection. These genes have the potential to assist the development of new coffee rust control strategies. Collectively, our results provide understanding of expression profiles in coffee-H. vastatrix interaction over a time course in susceptible and resistant coffee plants.

Introducción. El control de enfermedades en diversos cultivos se hace en su mayoría por métodos de origen químico, los cuales para productores con cultivos de pequeña escala comercial resultan muy costos y por ende poco asequibles. El control biológico es una alternativa que puede ayudar en cuanto a la reducción de gastos y sostenibilidad ambiental, a su vez se hace uso de elementos propios o extraídos del mismo cultivo, como en este caso la cascara del cacao que se desperdicia normalmente. Objetivo. El objetivo de este estudio fue evaluar la fracción sana de mazorca y rendimiento después de la aplicación de dos tratamientos uno biológico y otro de síntesis química. Materiales y métodos. Este estudio se realizó en Santo Domingo, Antioquía (COL), en una finca de un pequeño productor de cacao, donde se seleccionaron 17 árboles en 3 lotes, se aplicó un control biológico correspondiente a un lixiviado combinado con el hongo Trichoderma spp., asperjado al árbol (n=6). Un tratamiento de síntesis química, inductor de resistencia (K 3 PO 3 ) inyectado en el tronco del árbol (n=5) y un control sin aplicación (n=6). Luego de la aplicación, se contaron las mazorcas sanas y enfermas y se registró el peso húmedo de las semillas durante un periodo de 2 meses en cada uno de los tratamientos. Resultados y conclusiones. No se encontró diferencia significativa en las fracciones de mazorcas sanas y el rendimiento, aun así, el control biológico mostró una mejora con relación al control, lo cual indica su potencial como una alternativa de bajo costo para plantaciones de pequeña escala.

Plants react to biotic and abiotic stresses with a variety of responses including the production of reactive oxygen species (ROS), which may result in programmed cell death (PCD). The mechanisms underlying ROS production and PCD have not been well studied in microalgae. Here, we analyzed ROS accumulation, biomass accumulation, and hydrocarbon production in the colony-forming green microalga Botryococcus braunii in response to several stress inducers such as NaCl, NaHCO3, salicylic acid (SA), methyl jasmonate, and acetic acid. We also identified and cloned a single cDNA for the B. braunii ortholog of the Arabidopsis gene defender against cell death 1 (DAD1), a gene that is directly involved in PCD regulation. The function of B. braunii DAD1 was assessed by a complementation assay of the yeast knockout line of the DAD1 ortholog, oligosaccharyl transferase 2. Additionally, we found that DAD1 transcription was induced in response to SA at short times. These results suggest that B. braunii responds to stresses by mechanisms similar to those in land plants and other organisms.
