ABSTRACT:The examination of caecal contents allows one to investigate the exposure of colon mucosa to dietary toxins and the chemical contaminants present in animal feed. The objective of the present study was to examine the influence of Fusarium mycotoxins (zearalenone, ZEN, and deoxynivalenol, DON), administered separately and as a mixture, on the genotoxicity of caecal water (CW) from gilts fed these mycotoxins. CW genotoxicity was evaluated with the comet assay using the LLC-PK1 porcine epithelial kidney cell line. It was shown that after the first week of the experiment, the presence of DON in animal feed led to increased CW genotoxicity in the proximal colon, while the presence of DON and ZEN + DON had a similar effect in the distal colon (ANOVA, P < 0.05). After the third week of experiment, elevated genotoxicity in the distal colon was observed for all experimental groups of gilts, and it was 62%, 52.4%, and 52.8% higher for ZEN-, DON-, and ZEN + DON-fed animals, respectively, than for control group animals. However, no effect on CW genotoxicity in the proximal colon was seen. After six weeks, the presence of ZEN in the proximal colon increased CW genotoxicity by 103% in comparison with the control group. In the distal colon, after the sixth week, CW genotoxicity in all groups of animals fed with mycotoxin was significantly (by 80% to 116%) higher than in the control group. ZEN and DON administered as a mixture did not lead to an increased genotoxicity compared to either agent administered separately. Generally, the mycotoxins clearly started to increase the genotoxicity of CW from the third week of administration.

The study evaluated the proliferative activity of immunocompetent cells in the jejunal and iliac lymph nodes of prepubertal female wild boars exposed to deoxynivalenol and zearalenone in naturally contaminated feed. The evaluation was performed with the use of the MTT assay and 2 mitogens: lipopolysaccharide (LPS) and concanavalin A. Intensified proliferative processes in T and B lymphocytes were revealed. The mitogenic activity of LPS was more expressed in the lymphocytes of both iliac and jejunal lymph nodes in comparison with the control group. Proliferative activity was higher in iliac lymph nodes than in jejunal lymph nodes. A reverse trend was observed in the percentage of live cells, which was higher in jejunal lymph nodes during the evaluation of lymphocyte proliferation.

Środki spożywcze i materiały paszowe pochodzenia roślinnego są bardzo często wektorami niskich dawek mikotoksyn fuzaryjnych (28). Ze względu na szeroki zakres ich zastosowań w przemyśle spożywczym i w produkcji pasz, a także wysoki udział produktów zbożowych w diecie człowieka i zwierząt są ważnym elementem odpowiedzialnym za stan zdrowia ludzi oraz wydajność procesu produkcyjnego zwierząt (10). Najczęściej spotykanymi mikotoksynami fuzaryjnymi są zearalenon (ZEN) oraz deoksyniwalenol (DON).Zearalenon i jego metabolity powodują zaburzenia w rozrodzie zwierząt, ponieważ w swej aktywności są podobne do estradiolu (40). α-ZEL jest dominującym metabolitem ZEN występującym w organizmie świń, podczas gdy u innych gatunków zwierząt, jak np. u brojlerów, krów i owiec dominującym produktem metabolizmu ZEN jest β-ZEL o dużo mniejszej aktywności metabolicznej (9). Efekt aktywności ZEN zależy od nasilenia i charakteru procesów metabolicznych mających miejsce w organizmie zwierząt (18). Zależy również od statusu immunologicznego (5, 20) układu rozrodczego, który podlega ciągłym zmianom na skutek zmian hormonalnych wynikających z okresu dojrzewania płciowego, etapu cyklu rozrodczego czy ciąży. Wykazano, że ZEN może być immunotoksyczny w niskich stężeniach (33). W odpowiedzi na obecność mikotoksyny, wrażliwe komórki przechodzą wiele zmian w zakresie istotnych procesów metabolicznych (proliferacji i różnicowania się komórek, apoptozy, syntezy cząsteczek biologicznie aktywnych, itp.) (37). Pomimo że badania nad ZEN zostały ostatnio zintensyfikowane, to istnieje niewiele badań nad jego wpływem na barierę jelitową i potencjalnego związku pomiędzy ekspozycją na ZEN a stanami zapalnymi jelit (13).
