Laboratory experiments were performed to determine the antioxidant responses to nine phthalates (PAEs) in the liver of the goldfish Carassius auratus. The fish were injected with 10 mg/kg body weight of each PAE for 1 day and 4, 8, and 15 days. The potential biotoxicity of the PAEs were examined using the antioxidase and lipid peroxide indices. We determined that the superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and malondialdehyde (MDA) levels displayed different trends following prolonged treatment, suggesting that metabolism generated either less toxic or more active substances. Based on the intensity of enzymes inhibition, MDA content, and the calculated integrated biomarker response (IBR), the toxicity order was determined as follows: dibutyl phthalate (DBP) > diethyl phthalate (DEP) > diisodecyl phthalate (DIDP) > diphenyl phthalate (DPP) > butyl benzyl phthalate (BBP) > diallyl phthalate (DAP) > dicyclohexyl phthalate (DCHP) > dimethyl phthalate (DMP) > di(2-ethylhexyl) phthalate (DEHP). In particular, DBP, which exhibited significant inhibition of enzyme activity and the greatest decrease in MDA content, may be a highly toxic contaminant. Furthermore, our results suggest that the IBR may be a general marker of pollution.

Plastic waste reprocessing for circular economy: A systematic review of risks to occupational and public health from legacy substances and extrusion

Increasing aspirations to develop a circular economy for waste plastics will result in an expansion of the global plastics reprocessing sector over the coming decades. Here we focus on two critical challenges within the value chain that as a result of such increased circularity may exacerbate existing issues for occupational and public health (1): Legacy contamination in secondary plastics, addressing the risk of materials and substances being inherited from the previous use and carried through into new products when the material enters its subsequent use phase; and challenge (2): Extrusion of secondary plastics in reprocessing, an end process of conventional mechanical recycling of plastics, involving heating secondary plastics under pressure until they melt and can be formed into new products. Via a systematic review (PRISMA guidelines, adapted), we considered over 4,000 sources of information, refined and consolidated into 20 relevant sources, which were critically assessed. We also derive prevalent risk scenarios of hazard-pathway-receptor combinations, subsequently being ranked. Our critical analysis highlights that despite stringent regulation, industrial diligence and enforcement, occasionally small amounts of potentially hazardous substances are able to pass through these safeguards and re-enter in the new product cycle. Although many are present at concentrations unlikely to pose a serious and imminent threat, their existence may be an indication of a wider or possibly increasing challenge of pollution dispersion, as the plastics reprocessing sector proliferates. But, in the Global South context, such controls may not be in place. Several studies showed emission control by passive ventilation, through open doors and windows followed by dilution and dispersion in the atmosphere, resulting in increased occupational exposure. It is recommended that further investigations are undertaken to establish the scale and magnitude of such phenomena, especially given the limited evidence base, with results informing improved future risk management protocols of a circular economy for plastics.

Influence of Phthalates on Cytokine Production in Monocytes and Macrophages: A Systematic Review of Experimental Trials

BackgroundPhthalates are a group of endocrine disrupting chemicals suspected to influence the immune system. The aim of this systematic review is to summarise the present knowledge on the influence of phthalates on monocyte and macrophage production and secretion of cytokines, an influence which could affect both pro- and anti-inflammatory abilities of these cells.Strategy and ResultsA systematic search was performed in Medline, Embase and Toxline in June 2013, last updated 3rd of August 2014. Criteria used to select studies were described and published beforehand online on Prospero (http://www.crd.york.ac.uk/NIHR_PROSPERO, registration number CRD42013004236). In vivo, ex vivo and in vitro studies investigating the influence of phthalates on cytokine mRNA expression and cytokine secretion in animals and humans were included. A total of 11 reports, containing 12 studies, were found eligible for inclusion. In these, a total of four different phthalate diesters, six primary metabolites (phthalate monoesters) and seven different cytokines were investigated. Though all studies varied greatly in study design and species sources, four out of five studies that investigated di-2-ethylhexyl phthalate found an increased tumour necrosis factor-α secretion/production from monocytes or macrophages. A summary of cytokine measurements was not possible since few studies were comparable in study design and due to insufficient reporting of raw data for most of the included studies.ConclusionResults from this review have suggested that at least one phthalate (di-2-ethylhexyl phthalate) has the ability to enhance tumour necrosis factor-α production/secretion from monocytes/macrophages in vitro, but also observed ex vivo. Influence of other phthalates on other cytokines has only been investigated in few studies. Thus, in vitro studies on primary human monocytes/macrophages as well as more in vivo studies are needed to confirm or dispute these findings.
