Increased Th1/Th2 (IFN-.GAMMA./IL-4) Cytokine mRNA Ratio of Rat Embryos in the Pregnant Mouse Uterus

Abstract: Abstract. Somatic cell nuclei can be dedifferentiated in ooplasm from another species, and interspecies cloned embryos can be implanted into the uteri of surrogates. However, no full pregnancies have been achieved through interspecific mammalian cloning. Rat blastocysts transferred into mouse uteri provide a unique model for studying the causes of interspecific pregnancy failure. In this study, intraspecific pregnancy (mouse-mouse) and interspecific pregnancy (rat-mouse) models were established. On Day 9 of pr… Show more

“…The ratio of IL‐4 (Th2‐type response) to IFN‐γ (Th1‐type response) is usually used to analyse the Th2 or Th1 bias of a generated immune response (Nan et al . ). In this study, the ratio IL‐4/IFN‐γ was 1·18 (Fig.…”

“…It can selectively inhibit the generation of IFN-γ and IL-2, therefore reducing the activity of large phagocytes, NK cells and LAK cells and resulting in the decreased cell-mediated immune. And thus, endometrial immune rejection is inhibited, immune tolerance is induced and fetal implantation gets uneventful [13][14] . In addition, a lot of steroid hormones excreted by the mother during the pregnancy, including estrogen, progesterone, chorionic gonadotropin and prostaglandin, take part in the regulation of the immune responses.…”

“…Cytokines are important mediators in the gestational process, which lead to a successful pregnancy since they are involved in the implantation of the blastocyst, embryo formation and development, and especially in the development of the central nervous system [46]. The uterus promotes an immune adaptation during pregnancy to prevent maternal rejection [47]. The IL-10 cytokine seems to be the key to maintaining the gestational process due to this protection effect of the fetal-placental unit, inhibiting the secretion of inflammatory cytokines, such as IL-6 and INF-γ [48].…”
