Increased serum levels of solublel-selectin (CD62L) in patients with active systemic lupus erythematosus (SLE)

Abstract: SUMMARYThe adhesion molecule L-selectin (CD62L) mediates lymphocyte recirculation and leucocyte rolling on vascular endothelium at sites of inflammation. Serum levels of soluble L-selectin (sL-selectin) were measured in patients with SLE in order to relate these levels to clinical activity and immunological parameters. An ELISA was used to detect the soluble form of human L-selectin (CD62L) in 42 patients with SLE and in 33 healthy individuals. The mean Ϯ s.e.m. values of sL-selectin were 1285 Ϯ 121 ng/ml for … Show more

“…The LAM1-102 mAb identifies an epitope within the epidermal growth factor/short consensus repeat regions of L-selectin, whereas the LAM1-116 mAb binds an epitope located within the lectin domain (5). Comparable results were obtained when the LAM1-101 mAb replaced the LAM1-102 mAb, which binds to a similar region of L-selectin (data not shown) (45). The presence of sL-selectin was readily detected in serum from normal adult B6 mice, with a mean concentration of 1.7 Ϯ 0.1 g/ml (Fig.…”

“…In the current study, mouse serum sL-selectin levels were quantified using a previously described human serum standard that contained a known concentration of sL-selectin (26,28). Others have used recombinant human L-selectin as a standard for measuring human sLselectin levels (45). Therefore, it is likely that humans and mice have similar, high levels of serum sL-selectin that are likely to influence leukocyte migration.…”

“…These cells interact via cutaneous lymphocyte associated antigen (CLA) on their surface with E-selectin molecules on dermal microvascular endothelial cells. Levels of soluble E-selectin correlate with active disease and levels of dsDNA antibodies in patients with SLE (248). E-selectin can be upregulated by UVB (249) and its expression is increased in CCLE and SLE photoinduced lesions (250).…”

“…Small-scale studies in lupus patients indicate that levels of soluble and membrane-bound E-selectin 9,10 and soluble L-selectin (sL-selectin) may be associated with SLE and disease activity. 11,12 The sL-selectin is derived from calcium-dependent endoproteolytic cleaving of membrane-bound protein from activated lymphocytes and from rolling neutrophils. 13,14 sL-selectin is present at high concentrations in the serum of healthy humans (B1.0 mg/ml) and retains functional activity perhaps inhibiting interactions involving its cell surface counterpart.…”
