In vivo study of hot compressing molded 50:50 poly (DL‐lactide‐co‐glycolide) antibiotic beads in rabbits

Abstract: The authors investigated poly (DL-lactide-co-glycolide) beads as an antibiotic delivery system in vivo for the treatment of various surgical infections. In this study, the copolymer 50:50 poly (DL-lactide):co-glycolide was mixed with vancomycin powder and hot compressing molded at 55 "C to form 8 mm in diameter biodegradable antibiotic beads. The antibiotic beads were implanted in the distal femoral cavities of rabbits for in vivo investigation. The local concentration of vancomycin was well above the breakpoi… Show more

“…However, there has not been an animal model specific for detecting the sequential changes in local antibiotic concentration. The animal model used in this study was designed by the authors specifically for the investigation of local antibiotic release profiles from antibiotic carriers [45]. In the rabbit model, a granulated bony cavity at the distal femur was created for the implantation of antibiotic beads.…”

“…[7][8][9][10][11][12][13][14][15] Alginate, a very promising biodegradable material, [16][17][18][19] has been utilized as a support matrix for entrapping and culturing malignant cells and chondrocytes. Furthermore, previous in vitro studies have identified a stabilizing effect of alginate beads on hydrophilic b-lactam antibiotics and epithelial proliferation growth factors.…”

“…In general, this sustained release rate was due to the novel 3D hierarchically nanostructures with mesoporous structure. In clinical applications, the breakpoint sensitivity of vancomycin for Staphylococcus aureus was 5 µg/mL, respectively[21]. The vancomycin concentrations released from HAp microspheres during the research period was around 76-155µg/mL…”
