In vivo evidence of the targeting of cartilaginous tissue by pyridinium functionalized nanoparticles

Abstract: Ultrasmall gadolinium based particles have been functionalized with positively charged pyridinium quaternary ammonium and labelled with (111)In. Evidence of their active targeting properties towards proteoglycans has been demonstrated in vivo after intravenous injection into rats opening thus a route to cancer imaging and therapy.

“…2 In our study, since GBNs were not yet designed to specifically target HNCSS tumor cells, we preferred to use intra-tumoral injections to better control the local concentration of GBNs, even though an homogeneous distribution of GBNs in the tumor cannot be certain, leading to treatment variability, which may explain the variability in the GBN + 10 Gy group. Three recently published studies by Morlieras et al [41][42][43] reported GBNs functionalized with 3 different ligands targeting tumors rich in melanin, proteoglycans or αvβ3.…”

“…Such deconvolution approaches with ESI-MS measurements were found to be well-adapted for solving mass distributions of polydisperse gold nanoclusters 51,[160][161][162][163][164][165] as well as for ultrasmall Gd-based silica NPs. [166][167][168][169][170][171][172][173]…”

“…The nebulisation was performed with 100 MBq/3 mL of 99m Tc-DTPA and 60 MBq/3 mL of 111 In-SRP suspension. The chelation of 111 In on SRP was performed following the protocol developed by Morlieras et al 36 and modified for the purification process. Briefly, the nanoparticles were suspended in milliQ water at the concentration of 100 mg/ mL.…”
