In vivo distribution of 5-Fluorouracil after peritumoral implantation using a biodegradable micro-device in tumor-bearing mice

Abstract: A novel implantable micro-device was used for delivery of 5-Fluorouracil (5-Fu), which was often used in the treatment of various human malignancies. The biodegradable poly(lactic-co-glycolic) acid (PLGA) was used as material. The purpose of this study was to investigate the efficiency of delivery of 5-Fu to the tumor via this delivery system. The distribution characters of the 5-Fu in tumor, plasma, peritumoral tissue, liver and kidney were compared after peritumoral implantation of micro-device and intraperi… Show more

“…However, 5-Fu and curcumin have their shortcomings, such as toxicity and very low bioavailability, so much work was focused on the development of nano-carriers to overcome these short-comings. 35,36 It was reported that there was a synergism between 5-Fu and curcumin, 37 which was also testified in our study. The nano-carrier could increase the endocytosis of plain drugs, which would further enhance the effect of synergism between 5-Fu and curcumin.…”

“…Mice were prophylactically treated with 5-FU (10 mg/kg or 40 mg/kg) in combination with or without daptomycin (DAP). All drug concentrations were within the human therapeutic dosage range ( 41 ). In order to assess if the effect of 5-FU is mediated by inhibition of AI-2 production, we simultaneously administered synthetic AI-2 (DPD).…”

“…Advances in lithographic technology have enabled the fabrication of PLGA microdevices. 10,169,170 In one study a micromold was fabricated by casting polydimethylsiloxane (PDMS) onto a lithographically patterned SU-8 mold. The PDMS mold was used to fabricate 5-FU-loaded PLGA microdevices with honeycomb structures and 5-FU was injected as a fine powder into the chamber.…”

“…10 This group of researchers also conducted another study to observe the pharmacokinetics of 5-FU released from this device. 170 Mice administered with 5-FU via IP injection (7.2 mg/mice) had a peak plasma concentration (C max ) of 0.83 µg/ml and time to peak plasma concentration (T max ) at 0.5 hours. The plasma concentration was lower than the limit of detection (50 µg/ml) within 2 hours.…”
