In vivo confocal Raman spectroscopy and molecular dynamics analysis of penetration of retinyl acetate into stratum corneum

Abstract: Spectroscopy data indicate that retinyl acetate permeates into the stratum corneum. Molecular dynamics data showed that retinyl acetate permeates in the membrane model and that their final location is deep inside the lipid bilayer. We showed, for the first time, a correlation between Raman permeation data and computational data.

“…In addition, it is good for following up the penetration/permeation of APIs [114,115]. Nowadays, Raman microscopy is a powerful technique to properly understand skin structure and percutaneous drug delivery [111,[116][117][118].…”

“…Eklouh-Molinier [211] combined CRM and cutometric data into a Partial Least-Squares (PLS) model in an attempt to profile spectral markers associated with ageing, and used this to create a "biological age" chart that mapped skin age based on its molecular state rather than its chronological age. Besides SC thickness and water content, measurements of ceramides/fatty acids and NMF content are possible with CRM, and can be used to assess barrier function [215,216], as well as the penetration of sunscreens [217,218], cosmetic ingredients [219][220][221], and other substances [220,[222][223][224][225][226][227][228].…”

“…Different methods have been described for obtaining depth profiles of topically applied actives using CRS [ 8 , 9 ]. In skin in-vivo studies, in-line measurements have been widely described for monitoring the distribution of actives or excipients on human skin [ 10 , 11 ]. However, the use of human skin faces a lot of difficulties concerning accessibility due to ethnic problems and poor convenience.…”
