In Vitro Synergy of Ciprofloxacin and Gatifloxacin against Ciprofloxacin-Resistant Pseudomonas aeruginosa

Abstract: Multidrug-resistant Pseudomonas aeruginosa with combined decreased susceptibility to ceftazidime, ciprofloxacin, imipenem, and piperacillin is increasingly being found as a cause of nosocomial infections. It is important to look for combinations of drugs that might be synergistic. Ciprofloxacin resistance by P. aeruginosa is mediated in part by an efflux pump mechanism. Gatifloxacin, an 8-methoxyfluoroquinolone, inhibits a staphylococcal efflux pump. An earlier in vitro study using an Etest synergy method and … Show more

“…Each test was performed in triplicate. For the first clear well in each row of the microtitre plate containing all antimicrobial agents, the fractional inhibitory concentration (FIC) was calculated as follows: FIC of drug A5MIC drugA in combination/ MIC drugA alone, and FIC of drug B5MIC drugB in combination/ MIC drugB alone (Pankey & Ashcraft, 2005). The FIC index (FICI), calculated as the sum of each FIC, was interpreted as follows: FICI,0.5, synergy; 0.5¡FICI,1, partial synergy; FICI51, additive; 2¡FICI,4, indifferent; 4,FICI, antagonism (Odds, 2003).…”

“…First, the MIC for each antimicrobial was determined according to the guidelines from the Swedish Reference Group for Antibiotics (SRGA; http://www.srga.org). For the combination (synergy) testing, two methods, fixed ratio and MIC/ MIC ratio, were performed [15][16][17]. Briefly, in the fixed ratio method, an Etest strip of the first antimicrobial was placed on an agar plate, its exact location marked on the back of the plate, and incubated for 1 h at room temperature.…”

“…The potentiating power of the n-hexane fraction of P. biglobosa stem bark extract on selected antibiotics was assessed using the time-kill assay method as described by [34,35]. Conical flasks containing 50 mL of sterile nutrient broth dosed with n-hexane fraction, antibiotics and their combinations at 1/2 × MIC and 1 × MIC were inoculated with the standardised inoculum (~10 6 cfu/mL) to serve as test experimental setup.…”
