There are no data available regarding the bronchospasmolytic activity and toxicity of water extract of Coptosapelta flavescens Korth root (WECFR). Therefore, this study aimed to determine: a. the in vitro tracheospasmolytic effect of WECFR; b. safety of WECFR using the brine shrimp lethality test (BSLT) and immobilization of Daphnia magna larvae (IDL). In study, a guinea pig tracheal ring was contracted with methacholine and cumulative doses of WECFR solution were administered. A dose-response curve (DRC) was plotted of the percentage of the tracheal ring relaxation response. To test whether the relaxation mechanism occured through stimulation of beta2-adrenergic receptors, the tracheal ring was incubated with propranolol. Data analyzed using analysis of variance, showed that the DRC of WECFR was obtained with p < 0.05 compared to the DRC of control, indicating that WECFR had a tracheospasmolytic effect. The DRC for propranolol-blocked WECFR did not shift to the right compared to the DRC of WECFR, confirming that the relaxation mechanism did not occur through the beta2-adrenergic receptors. Study b. assessed safety using BSLT and IDL. After exposing the larvae to WECFR and control for 24 and 48 h, respectively, the number of dead larvae was counted. Data analyzed using Probit program, showed that the lethal dose 50 of WECFR towards Artemia salina and Daphnia magna larvae was > 1000 ppm, which means that it was not toxic. This studies demonstrate that WECFR exhibits tracheospasmolitic effect, but not through beta2-adrenergic receptors; WECFR is safe for Artemia salina and Daphnia magna larvae.

Objective: This study aimed to compare the bronchorelaxation effect of the methanol extract, hexane fraction, ethyl acetate fraction, and methanol fraction of Coptosapelta flavescens Korth. roots (CFRs) on the isolated bronchial rings of guinea pig. Methods: Research design was experimental in which a 4-mm long bronchus of a male guinea pig was kept in an isolated organ bath containing Krebs-Henseleit solution at 37°C, pH 7.4, and flooded with carbogen gas. The bronchial ring was precontracted with methacholine and then given six doses of C. flavescens Korth. root’s methanol extract (MECFR) solution cumulatively. The extract’s solvent 10% dimethyl sulfoxide ethanol was used as negative control. This procedure was then repeated using CFRs hexane fraction (HFCFR) solution, CFRs ethyl acetate fraction (EAFCFR), and CFRs methanol fraction (MFCFR). Results: The bronchorelaxation effect of MECFR, MFCFR, EAFCFR, and HFCFR is shown by their dose-response curves (DRCs) which are significantly different compared with the extracts solvent’s DRC. The maximal efficacy (Emax) of MFCFR was the same (p>0.05) as the MECFRs Emax, but the EAFCFRs and HFCFRs Emax were smaller (p<0.05) than the MECFRs Emax. Conclusion: The bronchorelaxation effect of the MECFR on the guinea pig’s bronchial ring is similar to the CFRs methanol fraction, and is stronger than the CFRs ethyl acetate fraction and CFRs hexane fraction.

The comprehension of some diseases at the cellular and tissue scale is fundamental for the development of effective treatments for such conditions, and in this regard, Raman micro‐spectoscopy, atomic force microscopy (AFM), and tip‐enhanced Raman scattering (TERS) are techniques that have featured prominently in the characterization of biological materials like cells and tissues, since they allow the acquisition of detailed physical, chemical, biophysical, and physiological properties of the surface of these kind of samples such as elasticity, viscoelasticity, wettability, topography, molecular composition, and others. Due to this, this literature review has as its primary scope the presentation of some of the recent advances and new analysis methodologies using Raman, AFM, and TERS, pointing out what they can offer toward the investigation of the behavior of cells and biological tissues. Limits of usage, drawbacks, and future perspectives are presented and discussed.
