The immune system acts as a defense mechanism against foreign antigens. Impairment of the immune system leads to the development of chronic diseases such as respiratory infections, cancer, cardiovascular diseases, and neurodegeneration. Macrophages, natural scavengers that are part of innate immunity, are known to directly participate in scavenging foreign antigens. The functional modulation of macrophages could be an effective treatment for pathogens. Seaweeds are marine macroalgae known to exhibit multiple bioactive properties. Thus, this study evaluated the immune-enhancing properties of marine brown algae extracts of Sargassum horneri (SH), Undaria pinnatifida (UP), and Sargassum fusiforme (SF) on murine macrophage cells. The results showed that all three algal extracts stimulated cell proliferation. SH and UP outshined SF in enhancing the expression levels of IL-1β, TNF-α, and IL-6 at almost all the concentrations tested as compared to SF which showed similar effects only at 200 or 400 μg/mL. A similar trend was seen in TNF-α, NO, and PGE2 production. Additionally, only SH and SF could enhance the mRNA expression levels of IL-12, and only SH upregulated the mRNA expression level of IL-10. The algal extracts also enhanced the phagocytosis activity of macrophages at 50–400 μg/mL for SH and 100–400 μg/mL for UP and SF. In conclusion, we found that these algal extracts could be considered immunomodulators that enhance the functional activity of macrophages.

Andrographis paniculata Burm. F Nees used to treat COVID-19 in several Asian countries because it has metabolites with pharmacological activity that may decrease the pathogenesis features of COVID-19. The extraction solvent significantly affected the composition and concentration of the metabolites. Therefore, the inhibitory effects of nitric oxide of A. paniculata extract based on differences in extraction solvent concentrations were investigare and also identified its metabolite profile. The leaves of A. paniculata were extracted with three different solvents like water solvent, 50% ethanol and pure ethanol. Nitrite oxide (NO) inhibitory activity on LPS-activated RAW 264.7 macrophage cells was used to evaluate the anti-inflammatory activity of the extract, the identification of metabolite profile using LC-MS/MS. Extract pure ethanol had the highest inhibition potential against NO at 91.02%, whereas water and 50% ethanol solvents had inhibitory potentials of 21.35% and -8.13%, respectively. The metabolites 14-deoxyandrographoside and 14-deoxy-17-hydroxyandrographolid were only identified in the pure ethanol extract. This study shows that differences in solvent concentrations play an essential role in enhancing the pharmacological potential of A. paniculata as an anti-inflammatory candidate for COVID-19 treatment.

Objective: Andrographis paniculata has long been a traditional medicinal plant in Indonesia. This study is intended to evaluate the anti-pro-inflammatory cytokines of 98% ethanol extract of A. paniculata by in vitro and in silico approaches. Inhibition of pro-inflammatory cytokines is also one of the therapies in treating COVID-19. Methods: The molecular docking approach was utilized as a first screening to evaluate the potential for suppression of macrophage cell activation; an ADMET prediction test was performed to determine the pharmacological, pharmacokinetic, and toxicity as a therapeutic target. TNF-α, IL-1ß, and IL-6 levels were measured using an ELISA method to investigate anti-cytokine pro-inflammatory activity in LPS-induced RAW 264.7 macrophage cells. LC-MS/MS was used to identify additional metabolite compounds. Results: Ethanol extract containing particular metabolites 14-Deoxyandrographoside and 14-Deoxy-17-hydroxyandrographolide inhibited TNF-α and IL-1β by 100% and IL-6 by 85.59%, respectively. While compared to the Dexamethasone molecule as a positive control, preliminary screening and ADMET prediction for the metabolite compound 14-Deoxyandrographoside exhibited relatively high binding stability to the CD14 receptor by -7.5 kcal/mol and was safe against various ADMET indications. Conclusions: This study reveals that the compound 14-Deoxyandrographoside in pure ethanol extract is a potential anticytokine agent candidate for treating pro-inflammatory cytokines, including COVID-19 infection.
