In vitro function of granulocytes isolated from blood of normal volunteers using continuous‐flow centrifugation in the IBM‐aminco celltrifuge and adhesion‐filtration leukapheresis using nylon fiber

Abstract: Granulocytes were harvested from each of five healthy male volunteers once by continuous flow centrifugation with the IBM-Aminco Celltrifuge, and once by adhesion filtration leukapheresis with nylon fiber. Granulocyte recovery and purity were significantly better with the filtration leukapheresis system than with continuous flow centrifugation. Measurements of trypan blue dye exclusion and muramidase activity were similar to those in control granulocytes regardless of the method of isolation. Granulocyte-stimu… Show more

“…Evaluating the functional capacities of PMNL one has to take into account, of course, the procedure by which the PMNL have been collected as well as the storage conditions. A marked functional impairment of degranulation, phagocytosis and killing of test-bacteria has been noted in PMNL-preparations collected by filtration leukapheresis [12,13]. Furthermore, the in vivo recovery of PMNL prepared by filtration leukapheresis seems to be impaired as compared to the recovery of PMNL procured by continuous flow centrifugation [14].…”
