In Vitro Evaluation of Enterococcus faecalis Adhesion on Various Endodontic Medicaments

Abstract: E. faecalis in endodontic infection represents a biofilm type of disease, which explains the bacteria’s resistance to various antimicrobial compounds and the subsequent failure after endodontic treatment. The purpose of this study was to compare antimicrobial activities and bacteria kinetic adhesion in vitro for three endodontic medicaments with a clinical isolate of E. faecalis. We devised a shake culture which contained the following intracanalar preparations: CPD, Endoidrox (EIX), PulpCanalSealer (PCS); the… Show more

“…1). The plaque and bleeding indices recorded on the incisor and the laren Protokolle wurden bereits in früheren Publikationen beschrieben [6,17] Bei der initialen Optimierung der Echtzeitreaktion wurden PCR-Produkte per Agarosegel und über Schmelzkurven analysiert, um eine korrekte Produktgröße der Proben zu gewährleisten. Die positiven Reaktionen zeigten TmSpitzen von 7-90°C.…”

“…The molecular protocols used in this study have been previously described [6,17]. Each paper point was suspended in 50 μl of pure dimethyl sulfoxide (DMSO) and centrifuged for 30 s; 2 µl of this was used as DNA suspension for realtime PCR reactions.…”

“…Genomic DNA was obtained by the Hexadecyl trimethyl‐Ammonium Bromide (CTAB) modified previously described method (Denotti and others ) using 400 μL of 0.9% Sodium Chloride containing 0.2 g of bottarga mixed with reference strains. Briefly, this suspension was combined with 70 μL of 10% sodium dodecyl sulphate (SDS) and 5 μL of proteinase K at a 10 mg/mL concentration (SIGMA‐Aldrich, St. Louis, Miss., U.S.A.).…”

“…After the incubation, the Aa was suspended in vials containing Schaedler broth to obtain a concentration with a turbidity equivalent to the No. 3 McFarland standard (about 10 8  CFU/mL) then diluted to 1/100 (obtaining a 10 6  CFU/mL) using a spectrophotometer at 620 nm (DMS100s, Varian, NH, USA) [19]. …”
