A reliable protocol for rapid proliferation of callus and multiple shoots formation from Solanum viarum leaf callus explants were experimentally demonstrated in our present investigation. Administration of 0.1 to 0.6 mg/L silver nitrate (AgNO3) in the Murashige and skoog media (M.S media) had shown significant difference in callus formation when compare to non-supplemented media. Dark green nodular organogenic callus with 100% regeneration was observed in the media administered with.0.4 mg/L AgNO3, 0.5mg/L BAP(Benzyle amino purine) and 0.5mg/L NAA(Napthalene acetic acid). Regenerative callus on 0.4mg/L AgNO3, 2.0 mg/L BAP and 0.5mg/L IAA(Indole acetic acid) M.S media consistency, responded the best with maximum mean number of shoots (19± 0.36) and maximum shoot length (7.57 ± 0.05 cm) was observed at 2.0 mg/L BAP and 0.5 NAA with very high regeneration frequency (95%). Rhizogenesis of in vitro raised shoots under the potential influence of AgNO3 (0.4 mg/L) was found to be maximum expressed in terms of number of roots (40.2 ± 0.13) in the media supplemented with 1mg/L IAA with 95% regeneration potency and the maximum root length was 11.28 ± 0.14cm at 2mg/L IAA. Our investigation could clearly contribute to the improvement in the micropropagation of medicinally important endangered plant Solanum viarum

A comparative growth study was conducted on juice vesicles cultured in the form of various fruit explant types (equatorially bissected fruit halves, longitudinally bissected fruit halves, one‐eighth sections of fruit, one‐quarter sections of fruit, whole carpel segments, 2 or 3 mm thick equatorial slices of fruit, and 1 cm2 fruit endocarp pieces) from 15 mm diam Citrus limon (L.) Burm. f. cv. Eureka lemons. Juice vesicles within equatorial fruit halves produced the least amount of callus. Furthermore, these juice vesicles grew similarly to juice vesicles occurring in the tree grown fruit. A study of cultured equatorial fruit halves using 10–45 mm diam lemons was then conducted. Fruit half cultures containing juice vesicles could be readily established from 15–45 mm diam lemons. Vesicles from 10 mm diam fruit halves, however, invariably produced callus. Vesicles cultured within fruit halves produced proportionately less callus as their fruit diam increased. Juice vesicles cultured in 15–30 mm diam fruits lost their original green color and turned opaque as they matured (i.e., after 3–6 months in culture). A method is also presented, whereby whole lemon fruits can be established and maintained in vitro. Lemons, 35–45 mm in diam, were the best explant sources for establishing whole fruit cultures. Juice vesicles in whole fruit cultures may remain viable for up to 8 months in culture.

Induction of Adventitious Branches From Cultured Citrus Juice Vesicles–a Potential Means of Proliferation

The noncallusing morphogenetic properties of Citrus juice vesicles cultured in vitro are unknown and were herein studied. Juice vesicles isolated from 120–180‐day‐old fruits are capable of proliferation via adventitious vesicle branching in vitro. Gibberellic acid levels of 1–100 mg/L greatly enhanced adventitious vesicle branching while delaying vesicle senescence as evidenced by the vesicles' ability to retain its green color for up to 4 to 6 mo in culture. Vesicles grown on media without gibberellic acid readily matured, turning opaque within 2 mo in culture and rarely produced adventitious branches. Additions of 0.1 mg/L α‐naphthaleneacetic acid or 1.0 mg/L benzylaminopurine to media containing 10 mg/L gibberellic acid further enhanced vesicle branching in some species and cultivars. Adventitious juice vesicles have been induced from a variety of citrus species, including Citrus grandis (L.) Osb., C. hystrix DC., C. limon (L.) Burm. f., C. medica L., C. paradisi Macf., C. reticulata Blanco, and C. sinensis (L.) Osb. Adventitious vesicle branches originate from primordia initiated on the surface of the cultured vesicle. These primordia commonly occurred on the terminal meristem region of the vesicle. However, some species (e.g., C. grandis and C. paradisi) produced adventitious vesicle branches from their bodies and stalks as well. Distinct vesicle branches begin to appear on preformed vesicles after 30–60 days in culture. A survey to determine the natural occurrence of vesicle branching in various Citrus species was also conducted. Natural vesicle branching commonly occurred in C. grandis, C. paradisi, and in some cultivars of C. reticulata, but was absent in most citrus species and cultivars (e.g., C. aurantifolia (Christm.) Swing., C. canaliculata Hort. ex Y. Tan., C. hystrix, C. limon, C. medica, C. sinensis). It was found that some species which failed to exhibit the adventitious branching phenomenon in nature did so in vitro (e.g., C. hystrix, C. medica, and C. sinensis).
