In Vitro Culture of Heuchera villosa ‘Caramel’

Abstract: The procedure for Heuchera villosa ‘Caramel’ propagation was investigated, which involves shoot regeneration, rooting of regenerated shoots, and acclimation of regenerated plantlets. Petioles, as explants, were cultured on MS medium supplemented with 1-naphthylacetic acid (NAA), benzylaminopurine (BA), thidiazuron (TDZ) and callus formed on all media. Shoots were observed to proliferate from callus on media with BA and NAA, whereas no shoots regenerated on media with TDZ and … Show more

“…Also the application of BA resulted in a high regeneration -over 90% for all the concentrations -but the results of rooting were less favourable than in the presence of mT. Comparable results were reported by Zhao et al (2017) for the micropropagation of Heuchera villosa 'Caramel' on MS containing NAA and BA or TDZ. The highest shoot regeneration (90.6%) was obtained for 0.1 mg•L -1 BA and 0.01 mg•L -1 NAA.…”

“…There were 16% more plants which survived relative to the untreated cuttings. Other authors who did not treat microcuttings with ABA reported comparable values: 85% of living plants for Heuchera villosa 'Caramel' (Zhao et al 2017) and 73% for Heuchera sanguine (Hosoki and Kajino 2003). It is thus obvious that even a low concentration (1%) of ABA can improve plant's adaptive ability and make microcuttings less sensitive to stress conditions and water loss.…”

“…Likewise, in plant cell, tissue and organ culture cytokinins are among the most important plant hormones for regulating growth and morphogenesis (van Staden et al, 2008). Among the main CKs used in tissue culture we have BAP, KIN, 2iP, ZEA and TDZ; for example, micropropagation by axillary budding of Quercus ilex was achieved by culturing shoots with zeatin (Martínez et al, 2017), in Phoenix dactylifera acclimatization was improved with KIN (Hassan, 2017), in Heuchera villosa 'Caramel', BA in combination with NAA the regenerated shoots grew normally, without hyperhydricity (Zhao et al, 2017). On the other hand, TDZ is a synthetic cytokinin that induces many responses in tissue culture such as stimuling the induction of callus and shoot regeneration in some plants species as Arachis hypogaea (Akasaka et al, 2000), and in Scutellaria bornmuelleri 0.5 to 4.0 mg L -1 TDZ induced efficient in vitro organogenesis and regeneration from leaf, stem and petiole explants (Gharari et al, 2019).…”

“…Fortunately, cutting and tissue culture are viable alternative ways to preserve distinctive phenotypic features and obtain numerous daughter plants. However, only a few varieties of Heuchera have been previously investigated in vitro, such as the cultivars Red Spangles [3], Caramel [4], Silver Scrolls [5], etc. And there are essentially no scienti c publications on its cutting propagation to date.…”
