In vitro conservation of Ceropegia elegans, an endemic plant of South India

Abstract: The purpose of this study was to develop in vitro techniques for conserving endemic species, Ceropegia elegans by axillary shoot multiplication. Murashige and Skoog (MS) medium with kinetin (Kn) 23.20 μM + indole-3-acetic acid (IAA) 5.71 μM was the best for axillary bud proliferation inducing a mean of 7.11 ± 0.07 shoots per node. Shoots developed were rooted best on half strength MS medium with 4.90 μM indole-3-butyric acid (IBA). Plantlets established in pots exhibited 72% survival rate.

“…attenuate [10] , Scrophularia takesimensis Nakai [4] , Sterculia urens Roxb [9] , and Gossypium hirsutum [5] . Contrary to the present findings, a high percentage of shoot induction (60%) has been reported at 24.6 µmol 2iP from nodal explants of Ceropegia elegans [11] . The lesser number of shoots induced by Kin in the present study also coincides with the reports in Caralluma adscendens [10] .…”

“…Cytokinin alone did not induce multiple shoot formation remarkably in S. tonkinensis. Induction of shoots was not stimulated by increasing the concentration (8)(9)(10)(11)(12)(13)(14)(15)(16) µmol ) of all the cytokinins tried.…”

“…In the present study medium supplemented with kinetin showed less number of shoots which is in agreement with some other cases like that of C. candelabrum [24] and C. attenuata [10]. Some other plant species also showed dependence on cytokinins viz; C. elegans [25], C. spiralis [26], Holostemma ada-kodien [27] and Hemidusmus indicus [28]. The average performance of TDZ was demonstrated in the present study as well as in C. thwaiitesii [29].…”

“…Interestingly, we could achieve the best organogenesis response (adventitious shoot regeneration and elongation) and produced 36 shoots per petiole explants grown in MS medium fortified with 2-iP (9.8 μM) and NAA (10.7 μM). 2-iP was stated as a potent hormone in inducing maximum number of shoots in not only Bixa (D'Souza and Sharon 2001) but also in a wide range of plant species (Sergio et al 2000;Bhaskaran and Jayabalan 2005;Hiregoudar et al 2006;Sujatha and Ranjitha Kumari 2007;Ping Luo et al 2009;Sivanesan et al 2008;Hussain et al 2008;Aruna et al 2012;Krishnareddy and Pulliah 2012;Sonali et al 2013).…”
