In Vitro Comparison of Biocompatibility of Calcium Silicate-Based Root Canal Sealers

Abstract: The aim of this study was to assess the effect of three calcium silicate-based sealers (EndoSeal MTA, Nano-ceramic Sealer, and Wellroot ST) and two epoxy resin-based sealers (AH-Plus, AD Seal) on various aspects, such as cell viability, inflammatory response, and osteogenic potential, of human periodontal ligament stem cells (hPDLSCs). AH-Plus showed the lowest cell viability on hPDLSCs in all time periods in fresh media. In set media, hPDLSCs showed no significant differences in cell viability among all the t… Show more

“…Unsurprisingly, Bio-C Sealer and TotalFill BC Sealer were significantly higher than AH Plus. These results were in line with those of a previous study that showed that EndoSequence BC, BioRoot RCS, and Endoseal MTA showed a significant increase in mineralized nodule formation compared to the AH Plus group [11]. In addition, a recent study with endodontic sealers showed that TotalFill BC Sealer induced alkaline phosphatase activity and strong mineralization capacity [26].…”

“…Bio-C Sealer and TotalFill BC Sealer are bioactive in nature; i.e., they induce mineralization when in contact with biological tissues [20]. AH Plus was used as a control because it is one of the most commonly used and investigated root filling materials [11,21,22].…”

“…Calcium silicate-based sealers, including more reactive calcium-silicate-based bioactive glass, known as bioactive root canal sealers, have been recently introduced [ 30 , 31 , 32 ]. Bioactive glass has been mainly used for dental hard tissue regeneration; however, it has also recently shown promise for the repair of several complex tissues [ 33 ].…”

“…(Shashank et al, 2019) alka¬line pH could neutralize the lactic acid from osteoclasts and prevent dissolution of minera¬lized components of teeth. (poggio et al, 2017) We have numerous highly cited publications on well designed clinical trials and lab studies (Govindaraju, neelakantan and Gutmann, 2017;azeem and Sureshbabu, 2018;Jenarthanan and Subbarao, 2018;manohar and Sharma, 2018;nandakumar and nasim, 2018;Teja, Ramesh and priya, 2018;Janani and Sandhya, 2019;khandelwal and palanivelu, 2019;malli Sureshbabu et al, 2019;poorni, Srinivasan and nivedhitha, 2019;Rajakeerthi and ms, 2019;Rajendran et al, 2019;Ramarao and Sathyanarayanan, 2019;Siddique, nivedhitha and Jacob, 2019). This has provided the right platforms for us to pursue the current study.…”
