In vitro assessment of immunomodulating activity of the two Lactobacillus strains isolated from traditional fermented milk

Abstract: The objective of this study was to evaluate the in vitro immunomodulating capacity of Lactobacillus coryniformis subsp torquens T3L (L. coryniformis T3L) isolated from traditional fermented yak's milk in Tibet, China, and Lactobacillus paracasei supsp. paracasei M5L (L. paracasei M5L)isolated from kumiss in Sinkiang, China was used as control. The effects of live bacteria, cell wall and genomic DNA of the two Lactobacillus strains on human peripheral blood mononuclear cells (PBMCs) proliferation, production of… Show more

“…In this study, we used FISH probes to identify bacterial species already employed as probiotics for tilapia (Jatobá, Vieira, Buglione, Silva, Mouriño, Jerônimo, Dotta & Martins ; Zhou, Tian, Wang & Li ), like Bacillus and Lactobacillus brevis . Similarly, we have evaluated the presence of Lactobacillus collinoides , Lactobacillus farciminis and Lactobacillus coryniformis that have been reported as potential probiotics for non‐aquatic animals (Tamura, Hori & Nakagawa ; Grimoud, Durand, Courtin, Monsan, Ouarné, Theodorou & Roques ; Tuo, Zhang, Han, Du, Zhang, Yi, Zhang & Jiao ). On the other hand, the use of Vibrio and Pseudomonas fluorescens probes was chosen due to the pathogenic action of these species on tilapia (Flick ).…”

“…Ten-milliliter inoculums were centrifuged at 4000 r/min for 10 min, washed twice with 10 mL sterilizing saline (0.9%), then resuspended in the same buffer. One milliliter of the suspension was mixed with 9 mL stimulated gastric juice and incubated in a shaking incubator (37 °C, 150 r/min) for 3 h. After that, its colony forming units (CFU) was separately counted at 0 and 3 h. The survival rate (SR) was calculated using the following equation: SR (%) = ( N 3 / N 0 ) × 100, where SR is the survival rate of Lactobacillus and N 0 is the number of viable cells (CFU/mL) at 0 h and N 3 is the number of viable cells (CFU/mL) at 3 h [ 20 ].…”

“…torquens T3L and Lactobacillus paracasei subsp. paracasei M5L) isolated from traditionally fermented milk exhibit immunomodulating capacity [ 20 ]. Lactobacillus fermentum HY01 ( LF -HY01) was also isolated from traditionally fermented yak yoghurt.…”

“…The Lactobacillus paracasei subsp . paracasei M5 strain was obtained from traditional koumiss in Sinkiang, China [ 10 ], and was selected in this study on the basis of its high adherence to human colonic epithelial cells [ 16 ], immunomodulatory activity [ 17 ], and antiproliferative activity against human colon cancer cell line HT-29 [ 10 , 12 ]. The Lactobacillus paracasei subsp .…”
