In vitro antioxidative and anti-inflammatory effects of the compound K-rich fraction BIOGF1K, prepared from Panax ginseng

Abstract: BackgroundBIOGF1K, a compound K-rich fraction prepared from the root of Panax ginseng, is widely used for cosmetic purposes in Korea. We investigated the functional mechanisms of the anti-inflammatory and antioxidative activities of BIOGF1K by discovering target enzymes through various molecular studies.MethodsWe explored the inhibitory mechanisms of BIOGF1K using lipopolysaccharide-mediated inflammatory responses, reporter gene assays involving overexpression of toll-like receptor adaptor molecules, and immun… Show more

“…2.7 | NO production assay RAW264.7 cells were treated with escalating doses of Mc-eWE (0-400 μg/ml) for 24 hr, and NO levels released in the cell culture media were measured using Griess reagent, as previously reported (Guevara et al, 1998;Hossen et al, 2017). 2.8 | Measuring mRNA expression levels by quantitative real-time PCR RAW264.7 cells and splenocytes were treated with escalating doses of Mc-eWE (0-400 μg/ml) for 24 and 12 hr, respectively. Total RNA was extracted using TRI reagent®, and cDNA was synthesized from 1 μg of total RNA using reverse transcriptase, according to the manufacturer's instructions.…”

“…The RAW264.7 cells or peritoneal macrophages were pre-treated with Rb-ME or L-NAME or Prednisolone (Pred) for 30 min., and then stimulated with LPS for 24 h. The supernatant (100 µL) obtained was mixed with 100 µL of Griess reagent, as reported previously [18,19]. The absorbance of this mixture was measured at 540 nm and the concentration of NO was calculated by comparison with a standard curve.…”

“…In another study, the in vitro antioxidative and anti‐inflammatory effects of BIOGF1K, a compound K‐rich fraction isolated from the root of Panax g inseng, were accessed, being observed a significant suppression of IKKβ and TBK1, involved respectively, in the transcriptional regulation of NF‐κB and IRF3 . Moreover, the microbial transformation of ginsenoside Rb1, Re and Rg1 increased their anti‐inflammatory properties via lipopolysaccharide‐stimulated murine RAW 264.7 macrophages and even xylene‐induced acute inflammatory model of mouse ear oedema .…”
