A method for adventitious shoot regeneration from leaves of micropropagated peach shoots has been developed. Apices were excised from in vitro shoot cultures of a seed-derived (juvenile) genotype (P16Cl5) and mature genotypes (Babygold 6, 842 Standard, San Giorgio and Yumyeong). Apices were cultured 21 days in the dark on a medium supplemented with 6-benzyladenine and α-naphthaleneacetic acid and then transferred to an auxin-free medium and incubated in the light for 21 days. The first four apical leaves were excised from these apices and cultured in the same way. During the dark incubation period, leaves developed a callus at the petiolar base. Adventitious shoots appeared on this callus after transfer to auxin-free medium and culture under light conditions. The morphogenic ability of the callus was maintained for several months.

An improved method for adventitious regeneration from apricot leaves is described. The use of the ethylene inhibitors silver thiosulphate (30-60 micro M) or aminoethoxyvinylglycine (0.5 micro M) increased regeneration percentages in Helena and Canino apricot cultivars and also the consistency of results from different experiments. Use of "Pure Agar" also improved regeneration from Helena leaves as compared with agargel or agarose. Regeneration rates for Canino were dependent on the medium in which the shoots were micropropagated. When different antibiotics were tested for their influence on regeneration, the combination of cefotaxime (0.13 m M) plus vancomycin (0.63 m M), which efficiently controls Agrobacterium growth, also increased regeneration percentages in Helena two-fold but did not affect regeneration in Canino. Kanamycin, an antibiotic widely used for selection of nptII transformed cells, promoted more rapid regeneration and higher regeneration rates from Helena leaves when added at low concentrations (8.6 and 17.1 micro M). With this improved procedure, regeneration from apricot leaves has been increased more than 200% as compared with rates reported previously.

Plums arose in three distinct regions, European plums ( Prunus domestica and others) from southern Europe and Asia Minor, Japanese plums ( Prunus salicina and others) from China, and the American plums ( Prunus Americana and others) from North America. The two major commercial species of plum are P. domestica (primarily for the dried fruit industry) and P. salicina (primarily for the fresh fruit industry). Breeding efforts have resulted in many high quality fruit cultivars that have been adapted for various local environmental and disease pressures. At present though, there have been few successes in breeding for resistance to plum pox potyvirus, a virus that not only affects the marketability of the fruit crop but the health of the tree. A number of research groups have begun to develop the techniques necessary to introduce resistance to plum pox virus through genetic engineering. These efforts have resulted in “HoneySweet”, a prune plum ( P. domestica ) that has been found to be highly resistant to plum pox virus in field trials. Work is continuing on improving the techniques used to engineer a plum pox virus‐resistant plum as well as producing more cultivars of plum with the resistance.
