In situ analysis of nanoparticle soft corona and dynamic evolution

Abstract: How soft corona, the protein corona’s outer layer, contributes to biological identity of nanomaterials is largely because capturing protein composition of the soft corona in situ remains challenging. We herein develop an in situ Fishing method that can monitor the dynamic formation of protein corona on ultra-small chiral Cu2S nanoparticles (NPs) allowing us to directly separate and identify the corona protein composition. Our method detects spatiotemporal processes in the evolution of hard and soft coronas on … Show more

“…[ 198 ] Recently, a biolayer interferometry‐based fishing strategy was developed for in situ analysis of corona protein adsorption and dissociation processes. [ 199 ] In the context of tracking the distribution of nanoformulations in the immune system, in vivo imaging and inductively coupled plasma analysis in combination with fluorescence‐activated cell sorting technology offer quantitative approaches to analyze the accumulation of nanoformulations in specific subsets of immune cells in lymphoid organs. The activation status of immune cells induced by nanoformulations can be revealed by flow cytometry analysis.…”

“…For example, the composition of the soft corona of chiral cuprous sulfide (Cu 2 S) is chirality-dependent and correlates to its stereo-selective biodistribution 79 . Time-dependent changes to the chirality-mediated soft and hard corona compositions were correlated with decreased blood circulation and trafficking to the liver 79 . Silver NPs typically dissolve into biologically active and highly toxic Ag + ions 80 .…”

“…Wang et al . [ 184 , 185 ] developed a series of synchrotron radiation analytical techniques to elucidate the valence of metal elements in various conditions and illustrate the interaction between NPs and proteins [ 186 , 187 ]. Besides, they comprehensively provided quantitative methods to monitor the content and transformation of NPs in the blood and different tissues [ 188 , 189 ].…”
