In search of the holy replicator

Abstract: After 40 years of searching for the eukaryotic replicator sequence, it is time to abandon the concept of 'the' replicator as a single genetic entity. Here I propose a 'relaxed replicon model' in which a positive initiator-replicator interaction is facilitated by a combination of several complex features of chromatin. An important question for the future is whether the positions of replication origins are simply a passive result of local chromatin structure or are actively localized to coordinate replication wi… Show more

“…Instead, origin firing is likely dependent on stochastic initiation at dormant origins defined as such be their chromatin status (Gilbert 2004), despite the presence of some origins that are sequence specific (Huberman 1998). Stochastic initiation is organized by feedback regulation from active replicons, which is mediated by the sensing of ongoing replication (Ge et al 2007;Ibarra et al 2008).…”

“…During postembryonic development, ecdysone is synthesized in and released from the prothoracic gland (PG) 1 under the regulation of neuropeptides such as prothoracicotropic hormone (3,5,6). Stimulation of the PG by neuropeptides involves several signaling cascades, including a cAMP-dependent pathway, a classical mitogen-activated protein kinase cascade, and an S6 kinase pathway (3,5,6). These pathways control synthesis of ecdysteroids from dietary cholesterol or phytosterols via a series of hydroxylation steps (3,4,6,7).…”

“…In insects, molting and metamorphosis are elicited by critical titers of ecdysone and 20-hydroxyecdysone. “…Despite the conservation of ORC and other trans-acting factors required for the assembly of the prereplicative complex ( pre-RC) and loading of the Mcm2 -7 helicase at origins, no conserved cis-acting elements directing DNA replication have been identified in higher eukaryotes (Gilbert 2004). Unlike in S. cerevisiae, ORC purified from higher eukaryotes shows little or no sequence specificity in vitro (Vashee et al 2003;Remus et al 2004).…”
