In-depth mass spectrometric mapping of the human vitreous proteome

Abstract: Mapping of proteins involved in normal eye functions is a prerequisite to identify pathological changes during eye disease processes. We therefore analysed the proteome of human vitreous by applying in-depth proteomic screening technologies. For ethical reasons human vitreous samples were obtained by vitrectomy from “surrogate normal patients” with epiretinal gliosis that is considered to constitute only negligible pathological vitreoretinal changes. We applied different protein prefractionation strategies inc… Show more

“…Peptides derived from opticin, beta-crystallin B2, and alpha-crystallin B were detected and are unique to vitreous (not seen in CSF). 5 Because of the small volume of schisis fluid, a more targeted analysis was performed and identified approximately 80 proteins. Opticin, beta-crystallin B2, and a large number of other vitreous proteins were detected in the schisis fluid.…”

“…The human vitreous humor is an aqueous solution that fills the posterior compartment of the eye [23]. Although the substructures of the vitreous and their proteins have been investigated recently [24], studies have centered on using vitreous humor as a diagnostic tool for retinal pathologies.…”

“…The main barrier for therapeutic compounds after intravitreal injection is the vitreous itself. Despite its clear appearance, the vitreous contains a 3D meshwork of connected fibers of collagen, hyaluronan and other proteoglycans like heparin and chondroitin sulfate which gives it its jelly-like texture (104-106). This structure caninterfere with diffusion of particles both by steric hindrance and also because charged delivery vehicles can interact with the charged proteins in the vitreous.…”
