Improvement of seminal parameters with Prelox®: a randomized, double‐blind, placebo‐controlled, cross‐over trial

Abstract: In a randomly allocated, double-blind, placebo-controlled, cross-over design, 50 infertile patients were treated for 1 month with placebo or a combination of l-arginine aspartate and Pycnogenol (Prelox). Semen samples were examined at 4 week intervals according to WHO criteria. Treatment with Prelox increased significantly the semen volume, concentration of spermatozoa, percentage of motile spermatozoa and percentage of spermatozoa with normal morphology compared with placebo. The placebo had no influence on t… Show more

“…In that study the estimated intake of omega-3 fatty acids increased by about 700 mg, and intake of fruits and vegetables and therefore antioxidants was also higher. In a placebocontrolled trial of men who took pycnogenol, a more potent antioxidant, together with a moderate dose of L-arginine, the mean IIEF score increased from about 15 to 27, suggesting that combining two agents known to stimulate NO may be more efficacious (37). There have been no studies of ED combining some or all of the factors discussed above that stimulate NO.…”

“…In a further randomized, double-blind, placebo-controlled, crossover trial of 50 men with at least 2 years of infertility, those same two agents were also shown to significantly increase semen volume, sperm density, percent motile sperm, and percent normal morphology (Fig. 4) (37). Such a global improvement in testicular function suggests an effect of increased blood flow rather than direct effects of one or both agents on diverse testicular tissues.…”

“…In regards to semen or sperm volume, it is indicated that L-Arg aspartate significantly increased sperm volume, concentration and motility through the antioxidative impact [16]. It has been confirmed that L-Arg enhances the rate of glycolysis, resulting in higher rates of adenosine triphosphate and lactate generation in spermatozoa [7].…”

“…This finding implies the involvement of NO in the observed effects of the drug. It is well-known that NO and its precursors increase the motility of spermatozoa by increasing energy production originating in the mitochondrial compartment (8) and that NO regulates various mitochondrial functions (1). Thus, in future studies, designed to define the precise mechanism behind the increase in the population of MT-G-positive sperm cells and motile spermatozoa, potential NO involvement warrants further consideration.…”
