Improved Skin Penetration Using In Situ Nanoparticulate Diclofenac Diethylamine in Hydrogel Systems: In Vitro and In Vivo Studies

Abstract: Abstract. Delivering diclofenac diethylamine transdermally by means of a hydrogel is an approach to reduce or avoid systemic toxicity of the drug while providing local action for a prolonged period. In the present investigation, a process was developed to produce nanosize particles (about 10 nm) of diclofenac diethylamine in situ during the development of hydrogel, using simple mixing technique. Hydrogel was developed with polyvinyl alcohol (PVA) (5.8% w/w) and carbopol 71G (1.5% w/w). The formulations were ev… Show more

“…The treatment was based on the “Guide for the development and submission of stability studies of chemically synthesized drugs” of the Ministry of Health and Social Protection of Colombia [ 69 ]. This was performed for the short-term under accelerated stability conditions at 40 °C and 75% relative humidity (RH) for one month [ 70 ]. Particle size analysis, TPA, and rheological behavior were measured at 7, 14, and 28 days of aging in the stability chamber compared with a freshly prepared hydrogel sample.…”

“…IAEC/DRL/05/2016). The required amount of collagen, PVA, AgSD, and bFGF were added in phosphate buffer solution (PBS, pH-7.4) followed by thorough mixing with the help of magnetic stirrer 49,50 . The temperature was maintained at 4ºC during the preparation and the pH of the formulation was adjusted to 5.0.…”

“…The surface tension of the formulation was determined by using a tensiometer CSC-Dunouy No. 70545 (Sengupta et al, 2016). A scaled-needle was positioned at zero before measurement then the ring balance was set by positioning balance disc forward or backward until the ring handle positioned at zero.…”
