Improved Micropropagation of Plantlets from Nodal Explants of Anchote (Coccinia abyssinica)—A Calcium- and Protein-rich Tuber

Abstract: The effect of phytohormones on breaking of dormancy of axillary buds in anchote and their subsequent shoot proliferation were examined. Anchote is an annual trailing endemic plant with high calcium content grown for its edible tuberous roots in Ethiopia. Nodal explants harvested from the greenhouse were sterilized using various concentrations of a commercial bleach (JIK) which contains 3.85% sodium hypochlorite (NaOCl) and time duration. The highest (85%) clean explants were obtained when 5% JIK was us… Show more

“…During the current study, kinetin was found to be more effective than TDZ in inducing somatic embryogenesis. These results are contrary to many studies which indicate that TDZ (a phenylurea type) is superior to the adenine types of cytokinins in inducing somatic embryogenesis in several plant species (Aboshama, 2011;Feng and Chen, 2014;Gill and Saxena, 1992;Kahia et al, 2016;Visser et al, 1992). In cacao, TDZ has been used by many workers (Ajijah et al, 2014;Garcia et al, 2016;Li et al, 1998;Maximova et al, 2002;Quainoo and Dwomo, 2012).…”

“…The work being reported is similar to that of Mayati (2015) who found that kinetin was better than zeatin in somatic embryogenesis of Hevea brasiliensis RRIM 2025. However, it conflicts with the work of Chen and Chang (2001), Wongtiem et al (2011), and Kahia et al (2016) who reported that kinetin was inferior to other cytokinins evaluated for the induction of somatic embryos in Oncidium 'Gower Ramsey', Manihot esculenta Crantz, and Coffea Arabica, respectively. During the current study, increasing the concentration of kinetin was found to inhibit induction of somatic embryos.…”

“…In the work on date palm, it was found that the shoot multiplication rate increased with increasing BAP concentration, but more hyperhydricity was observed (Mazri, 2015). Another work on anchote also indicated that hyperhydric shoots were observed on media supplemented with high concentrations of BAP and kinetin (Kahia et al, 2016). Similarly, our work suggested that shoots regenerated normally on media with lower concentration of BA (0.1 mg • L -1 ), whereas hyperhydricity phenomenon appeared on media with higher concentration of BA (0.1 or 0.5 mg • L -1 ) (Fig.…”

“…hyperhydricity induction, whereas the concentration has little influence (Kadota M and Niimi, 2003), in which synthetic phenylurea derivatives [N-(2-chloro-4-pyridyl)-N#-phenylurea (CPPU) and TDZ] produce more hyperhydric shoots than adenine derivatives (BA and kinetin). But in anchote, hyperhydrated shoots appeared on media supplemented with BAP and kinetin but not on TDZ media (Kahia et al, 2016). Therefore, exogenous cytokinin type and concentration suitable for shoot regeneration may depend on plant species.…”

“…Despite the importance of anchote, generally few studies have been conducted and almost nothing has been done with regard to improving the species through biotechnological applications before our first report on in vitro propagation (Yambo & Feyissa 2013) and molecular genetic diversity study using markers generated by Inter Simple Sequence Repeat (ISSR) techniques (Bekele et al 2014), which has been followed by others (Bekele et al 2013;Guma et al 2015;Kahia et al 2016;Abate et al 2019;Mekbib et al 2018). The objective of this article is to review research on anchote, present it in a comprehensive way, and suggest future directions of its improvement using biotechnological tools.…”
