Impedimetric Immunosensor for Detection of Plum Pox Virus in Plant Extracts

Abstract: This paper concerns the development of an immunosensor for detection of Plum pox virus in plant extracts. The sensor was based on gold electrodes modified with: 1,6-hexanedithiol, gold nanoparticles, anti-PPV IgG polyclonal antibody and BSA. It was used for determination of the virus in extracts from plum (Prunus domestica) and tobacco (Nicotiana benthamiana) leaves. The proposed electrochemical impedance spectroscopy (EIS) immunosensor displayed a very good detection limit of 10 pg/mL and a wide dynamic range… Show more

“…One of the most recent technique is impedimetric immunosensor, which is based on gold electrodes modified with 1,6-hexanedithiol, gold nanoparticles, anti-PPV IgG polyclonal antibody and bovine serum albumin. The proposed technique displays very good detection limit (10 pg/ml) and is able to differentiate the samples from healthy plants and the samples containing 0.01 % of infected plant extract [378]. The PCR-ELISA, respectively RT-PCR-ELISA, which is based on immunoenzymatic detection of PCR products, represents next possibility of ELISA/PCR modification with significant increase of sensitivity compared to both ELISA and PCR methods [374,379,380].…”

“…The immunosensor presented displayed better detection limit in the comparison to those already prepared in our laboratory (Table 1) (Jarocka et al, 2013(Jarocka et al, , 2011. The main reason for this could be the different way of immobilization procedures of sensing elements on the electrode surface.…”

“…Thus, the antibodies orientation on the electrode surface is rather random (Jarocka et al, 2011). For recognitions of Prunus Necrotic Ringspot Virus (PNRV), the antibodies were attached to via protein A.…”

“…The most popular mode for analyzing electrochemical impedance data is the plot of the imaginary impedance component versus real impedance component at each excitation frequency the Nyquist plot . Numerous impedimetric immunosensors have been already reported [7,30,31,38]. This approach has been successfully applied for detection of fragments of H5 hemagglutinin from avian inluenza viruses [9,10].…”
