Impaired long contact white matter fibers integrity is related to depression in Parkinson's disease

Abstract: This study suggested depression in PD was related to impaired white matter integrity especially the long contact fibers in the left hemisphere. These findings may be helpful for further understanding the potential mechanisms underlying depression in PD.

“…In addition to reduced FA in the genu and body of corpus callosum, left cingulum, and bilateral anterior and left superior corona radiata seen in PD subjects with apathy . Studies investigating depression in PD noted a reduction of FA in WM tracts of left anterior thalamic radiation, uncinate, and left anterior corona radiata, among others …”

“…14 Studies investigating depression in PD noted a reduction of FA in WM tracts of left anterior thalamic radiation, uncinate, and left anterior corona radiata, among others. 38 The differentiation of clinical symptoms was also observed in a study using PLS in order to relate brain atrophy with a range of clinical measures. 39 Interestingly, they found three distinct patterns of clinical presentations of PD, each related to a distinct atrophy pattern of gray matter.…”

“…Parkinson's disease (PD) is a progressive degenerative CNS disorder that affects the motor system. It is characterized by a profound degeneration of dopaminergic neurons in the brain, accompanied by chronic neuroinflammation, mitochondrial dysfunction, and widespread accumulation of α‐synuclein‐rich protein in Lewy's bodies . It has been proved that the α‐synuclein is a potent stimulator of inflammasome.…”

“…It is characterized by a profound degeneration of dopaminergic neurons in the brain, accompanied by chronic neuroinflammation, mitochondrial dysfunction, and widespread accumulation of α-synuclein-rich protein in Lewy's bodies. [43][44][45] It has been proved that the α-synuclein is a potent stimulator of inflammasome. Fibrillar α-synuclein in microglia leads to impairment of mitochondrial endocytosis and lysosomal dysfunction, which cause ROS dysregulation and activate inflammasome.…”

“…However, selective abnormalities in dopamine and other neurotransmitters in the limbic system were observed in PD patients with depression relative to PD patients without depression (12). Abnormalities in functional connectivity in the prefronto-limbic system (13) and structures in the prefrontal lobe, limbic system, and temporal lobe (14) were reported to be associated with depression in PD patients. Therefore, detecting the brain structural alterations in PD patients with depression may find markers for early diagnosis of PD.…”
