Allyl sulfur compounds play a major role in the chemoprevention against carcinogenesis. The present study compared the antiproliferative effects of diallyl sulfide (DAS), diallyl disulfide (DADS) and garlic extract on p53-wild type H460 and p53-null type H1299 non small cell lung cancer cells (NSCLC). The DAS and DADS treatment of both H460 and H1299 cells resulted in the highest numbers of cells in apoptotic state as measured by acridine orange staining, however, garlic extract treatment did not induce any significant apoptotic cells by MTT assay. DADS was found to be more effective in inducing apoptosis on NSCLC. The level of p53 protein in H460 cell was increased following DADS treatment. DAS and garlic extract treatment of H460 cells induced a rise in the level of Bax and a fall of Bcl-2 level. These results demonstrate that DAS, DADS and garlic extract are effective in reduction of anti-proliperative gene in NSCLC and suggest that modulation of apoptosis-associated cellular proteins by DAS, DADS and garlic extract may be the mechanism for apoptosis which merit further investigation as potential chemoprevention agents.

BackgroundThe resistance to chemotherapy is a major obstacle in the treatment of hepatocellular carcinoma (HCC), necessitating the discovery of additional agents. The use of natural products in this respect is extensively under investigation. Methods Two hundred and ten male albino rats were used, divided into 14 groups. Selected groups were pre-treated with vitamin C (ascorbic acid, AA) and/or diallyl sulphide (DAS). Hepatocellular dysplasia was initiated by a single intra-peritoneal (IP) injection of diethyl nitrosamine (DENA), diabetes was induced by a single IP injection of Streptozotocin (STZ). Other groups were treated with cisplatin (CP) alone or combined with AA and/or DAS for 14 weeks. This work aims to check if naturally occurring materials can help in improving response to CP chemotherapy using ploylol profile as a new index in management of HCC. ResultsThe results revealed that DENA significantly increased relative liver weight, serum ALT, AST and GGT activities, AFP, TNF-α and IL-6 levels, expression of aldose reductase (AR), sorbitol dehydrogenase (SDH) and Bcl2 proteins in the liver with decrease in body weight, expression of Bax protein and Bax/Bcl2 ratio in the liver. These effects were more pronounced in DENA+STZ group. These parameters showed relative correlation to AFP levels in HCC, in response to AA and/or DAS treatment except for SDH. Treatment with CP and/or AA and/or DAS significantly modulated most of these parameters except for SDH which showed no significant change in response to the suggested treatment. ConclusionsIn conclusion, AA and/or DAS showed apparent chemo-sensitizing, anti-inflammatory, AR inhibitory, apoptotic inducing and anti-diabetic activities, indicating new aspects for use as adjuvant to chemotherapy. Induction of diabetes in hepatocellular dysplasia -bearing rats showed higher resistance to chemotherapy. Ploylol profile is a useful prognostic tool in HCC patients with diabetic interference.

The Allium genus includes approximately 500 species, the most widely used of which are onions (Allium cepa), garlic (Allium sativum), leeks (Allium porrum), chives (Allium schoenoprasum), and shallots (Allium ascalonicum). Garlic is widely cultivated and consumed worldwide, and its beneficial effects have been known for thousands of years. It has been considered to increase longevity and to confer stamina and physical strength, and it has been used empirically as a vermifuge, antiseptic, antimicrobic, antipyretic, and analgesic. The scientific community has now become interested in the pharmacologic properties of Allium vegetables and their chemical constituents, particularly with regard to their effects on the cardiovascular system and in the prevention of cancer. Garlic has antihypertensive and antiarrythmic properties and exerts an antithrombotic effect through fibrinolytic activity and the reduction of platelet aggregation. Ingestion of garlic has also been reported to lower the concentration of triglycerides, cholesterol, and low-density lipoproteins and to increase the concentration of high-density lipoproteins in blood. These findings suggest that garlic has a preventive effect against atherosclerosis and its complications, including stroke, myocardial infarction, and thrombotic disorders. The findings in relation to cancer prevention that have been accumulated over the last 20 years are summarized here.We performed a systematic Medline search of the published literature on epidemiologic studies involving the relationship between consumption of Allium vegetables-garlic, onions, and leeks-and neoplasms/tumors. For the animal studies and for studies on mechanisms of action, we conducted searches for review articles. These lists of references were then updated using references from review papers and original research articles. Active Compounds in Allium VegetablesFresh garlic contains water, carbohydrates, proteins, fiber, and fat, as well as essential amino acids, vitamins, and minerals. When garlic is cut, chopped, or crushed, the clove's membrane is disrupted and S-allylcysteine sulfoxide (an odorless compound called alliine) is transformed enzymatically into allicin by alliinase (1). Allicin is responsible for the typical odor of garlic, but it is unstable and converts readily into mono-, di-, and trisulfides and other compounds such as ajoene. Onions mainly contain S-propenylcysteine sulfoxide (1), but also other sulfoxides, including S-propylcysteine sulfoxide and S-methylcysteine sulfoxide (2). S-Propenylcysteine sulfoxide, positional isomer of alliin, is called lacrimatory precursor, because it is tranformed by alliinase into the lacrimatory factor propanethial S-oxide. The lacrimatory factor is highly reactive and hydrolyzes to propionaldehyde, sulfuric acid, and hydrogen sulfide; it is also the precursor of several sulfur derivatives.
