Immunosensor Incorporating Anti-His (C-term) IgG F(ab’) Fragments Attached to Gold Nanorods for Detection of His-Tagged Proteins in Culture Medium

Abstract: Immunosensors based on gold electrodes (electrochemical) or gold discs (optical) modified with 1,6-hexanedithiol, gold nanorods and Anti-His (C-term) monoclonal antibody F(ab’) fragment are described. The antigen detected by the sensing platform is a recombinant histidine-tagged silk proteinase inhibitor (rSPI2-His6). Electrochemical impedance spectroscopy (EIS) and surface plasmon resonance (SPR) techniques were used as methods for detection of the antigen. This approach allows to detect the antigen protein i… Show more

“…The most popular mode for analyzing electrochemical impedance data is the plot of the imaginary impedance component versus real impedance component at each excitation frequency the Nyquist plot . Numerous impedimetric immunosensors have been already reported [7,30,31,38]. This approach has been successfully applied for detection of fragments of H5 hemagglutinin from avian inluenza viruses [9,10].…”

“…The presence of disulide or thiol group allow the F ab' 2 or F ab' fragments to covalent self-assembling on the gold nanoparticle surface [33,34]. This approach has been successfully applied for development of immunosensor destined for selective binding of antigen rSPI2-His present in the sample solution by F ab' fragment of antibody immobilized on a surface of the electrode using electrochemical impedance spectroscopy EIS and surface plasmon resonance SPR [30] as well as for selective detection of hemagglutinin from avian inluenza virus H5N1 [9,10]. The proteins " and G possessing high ainity toward the Fc part of immunoglobulins are frequently used for their covalent and oriented immobilization on the electrode surface [35].…”

“…Therefore, alternative sensors have been developed for specific detection of His-tagged proteins. They include gold nanorod sensors using surface plasmon resonance (Wasowicz et al 2008), impedance spectroscopy (Wasowicz et al 2010), and field effect transistors (Liu et al 2010). These detection systems are sufficiently sensitive (detection limits, *5-60 lg/ml), and fast (10-30 min), but these are based on nontraditional equipment for a molecular biology laboratory.…”
