Immunoreactive substance P is not part of the retinohypothalamic tract in the rat

Abstract: The retinohypothalamic tract (RHT) is a monosynaptic retinofugal pathway mediating information concerning the light/dark cycle from the retina to the brain's biological clock located in the suprachiasmatic nucleus (SCN). Light information, which daily adjusts (entrains) the rhythms of behaviour and physiology generated by the SCN, is mediated by two neurotransmitters, viz. glutamate and pituitary adenylate cyclase activating polypeptide (PACAP), co-stored in the RHT. Substance P (SP) modulates photic- and glut… Show more

“…Immunofluorescence procedures for double/triple antigen localization were performed as described previously (Hannibal & Fahrenkrug, 2002), using a mixture of the primary antibodies and biotinylated donkey antimouse antiserum (code no. 065‐151, Jackson Immunoresearch Laboratories, West Grove, PA, USA, diluted 1 : 800), Cy5‐conjugated donkey antirabbit antiserum (code no.…”

“…Although the exact mechanism by which a monophasic stimulus, light, produces either no response or a biphasic response in SCN neurons is unclear at present, Myers and co-workers [ 40 ] provided a potential molecular explanation for the phenomenon. Using electron microscopy and immunohistochemistry, they identified the excitatory amino acid glutamate and pituitary adenylate cyclase-activating polypeptide (PACAP) as the main neurotransmitters of the RHT [ 41 - 43 ]. Although substance P (SP) was thought of as a neurotransmitter of the RHT, there is now substantial evidence against this possibility [ 44 - 46 ].…”
