Immunohistochemical Expression of ABH/Lewis-Related Antigens in Primary Breast Carcinomas and Metastatic Lymph Node Lesions

Abstract: The expression of blood group antigens A, B, and H, as well as sialylated and nonsialylated forms of Lewis(a) and Lewis(x), was studied using immunohistochemical methods in normal and tumor tissues in the following cohort of patients: 51 patients with primary breast carcinoma, 13 with metastatic lymph node lesions, and 16 with benign tumors of the breast. As a control, normal tissue was obtained from a similar group of 22 patients with breast cancer. The noncancerous tissues expressed the same A/B/H antigens a… Show more

“…Like loss of ABO expression in tissue, [8][9][10][11] HER2/neu positivity is a result of altered glycosylation leading to HER2/neu expression. 12 Over-expression of HER2/neu cell membrane receptor protein in breast carcinoma is associated with tumor with the lack of correlation between blood type and HER2/ neu status noted in the present study.…”

“…Indirect immunoperoxidase staining was accomplished as follows [22]: monoclonal antibody TKH2 was diluted 1:10 in 0.05 M Tris, 2.5% bovine serum albumin, mouse liver acetone powder (100 mg/l; Sigma, St. Louis, Mo., USA) and 0.05% sodium azide. After treatment with 3% hydrogen peroxide in 0.05 M Tris-buffered saline (TBS; 50 m M Tris-HCl 200 m M NaCl, pH 7.6) for 5 min, the monoclonal antibody was incubated with tissue sections for 1 h at room temperature in a moist chamber.…”

“…Immunohistochemistry was performed as previously described, using an indirect immunoperoxidase staining technique (Nakagoe et al 1998). All steps were conducted at room temperature.…”

“…Tumors or tissues with more than 5% stained cells were de®ned as positively stained and those with 5% or less as negatively stained (Nakagoe et al 1998). Two of the authors judged the results of immunohistochemical staining independently, and the ®nal result was assigned by consensus.…”
