Immunohistochemical analysis of stromal fibrocytes and myofibroblasts to envision the invasion and lymph node metastasis in oral squamous cell carcinoma

Abstract: Background:Tumor cells work in close coordination with stromal elements from its stage of emergence to metastasis. The study was designed to assess the presence and distribution pattern of stromal fibrocytes and myofibroblasts in oral squamous cell carcinoma (OSCC). Possibility of using these stromal cells as a marker for invasion and lymphnode metastasis was evaluated.Materials and Methods:A total of 40 cases of OSCC consisting twenty cases of each lymph node positive (pN+) and lymph node negative (pN0) sampl… Show more

“…The cell migration and invasion assays were performed in the 24‐well Transwell plates (0.8‐m pore size filter; 3422; Corning) as follows: CALl27 and SCC15 cells were transfected with shNaa10p or IKKα siRNA for 24 hours with or without TGF‐β1 treatment, and 5 × 10 4 cells suspended in 200 µL serum‐free medium were added to the upper chamber. The lower chamber was filled with 800 µL DMEM culture medium containing 10% FBS (04‐001; Biological Industries, Kibbutz Beit HaEmek).…”

“…Oral cancer is one of the most common cancers in the world, and oral squamous cell carcinoma (OSCC) accounts for more than 90% of all oral malignancies. 1 , 2 , 3 , 4 Accumulating studies have shown that there is a close relation between the epithelial‐mesenchymal transition (EMT) and cancer progression. 5 , 6 , 7 EMT is the conversion of cells from epithelial phenotype to mesenchymal phenotype.…”

“…Oral cancer is one of the most common life-threatening types of cancer worldwide, with oral squamous cell carcinoma (OSCC) being the predominant form, accounting for over 90% of all oral malignancies [ 1 4 ]. OSCC is associated with high morbidity and mortality [ 5 , 6 ].…”

“…The clinical data of the specimens were classified by patient age, tumor size and degree of differentiation, which in turn was classified according to the Nottingham system ( Table I ) ( 23 ). Immunohistochemistry was carried out according to the EnVision two-step method as previously described ( 24 ), and PBS was used as a negative control. PGP9.5 was used as a neuronal marker, and its positive expression level was scored according to the following positive criteria: Cells that contain brown and yellow granules in the cytoplasm were considered positive, and the total number of positive cells in 100 cells in every five fields of view was counted.…”
