Immunogenicity and cytotoxicity of a platinum(iv) complex derived from capsaicin

Abstract: A platinum(iv) complex DCP acts as a chemoimmunotherapeutic agent and induces cancer cell death by damaging DNA, stimulating immunogenic response, and enhancing phagocytosis.

“…55 Mitochondrial membrane potential (MMP, Δ Ψ m ) has been recognized as a key indicator of mitochondrial function. 56 To establish whether apoptosis induced by complex 1 is related to mitochondrial dysfunction, MDA-MB-231 cells were treated with 1 at the indicated concentrations for 24 h and Δ Ψ m was evaluated by flow cytometry using a 5,5′,6,6′-tetrachloro-1,1′3,3′-tetraethylbenzimidazolycarbocyanine iodide (JC-1) kit. Cisplatin was used as a reference drug.…”

“…[62] In addition, capsaicin also has the effect of activating the immune system and reducing cancer immunosuppression. [63] According to the above studies, Sun et al [64] designed a Pt(IV) complex (Pt11; Figure 2) containing a capsaicin derivative as the axial ligand. Pt11 not only induces ICD in tumor cells, but also triggers an immune response by releasing the proinflammatory cytokines IFN-γ and TNF-α.…”

“…Apoptosis was previously thought to be tolerogenic, 34 but in certain cases, and more specifically in apoptotic cancer cells, it could activate the immune system. [35][36][37][38] This immunogenic apoptosis was associated with the release of HMGB1 and exposure of CRT, thus causing immune cell activation. [39][40][41] HMGB1 and ATP release have also been observed in necroptosis.…”
