Since current studies indicate possible infection of human lymphocytes with Chlamydia (Chlamydophila) pneumoniae, establishment of an in vitro C. pneumoniae infection model using lymphocyte cell lines was demonstrated. Human lymphoid cell lines (Molt 4 [T-cell] and P3HR1 [B-cell]) were utilized for this purpose besides human monocyte cell line (THP-1) and human epithelial cell line (HEp-2), as a reference of monocyte/macrophage cells and a positive control for support of C. pneumoniae growth, respectively. Both lymphoid cells (Molt 4 and P3HR1) supported the growth of C. pneumoniae as demonstrated by Chlamydia inclusion formation, detection of increased infective progenies and increased bacterial antigen levels. Similar data were obtained using monocyte THP-1 cells. However, the bacterial growth in these cells was less than that in HEp-2 cells. The electron microscopic study showed typical inclusions with many Chlamydia elementary bodies in lymphoid cells tested, similar to that seen in HEp-2 cells. These results indicate that C. pneumoniae can infect cells with lymphocyte properties and this infection model with lymphoid cell line cells could be valuable to study details of lymphocyte^C. pneumoniae interaction.

Epstein-Barr Virus Stimulates Torque Teno Virus Replication: A Possible Relationship to Multiple Sclerosis

Viral infections have been implicated in the pathogenesis of multiple sclerosis. Epstein-Barr virus (EBV) has frequently been investigated as a possible candidate and torque teno virus (TTV) has also been discussed in this context. Nevertheless, mechanistic aspects remain unresolved. We report viral replication, as measured by genome amplification, as well as quantitative PCR of two TTV-HD14 isolates isolated from multiple sclerosis brain in a series of EBV-positive and -negative lymphoblastoid and Burkitt's lymphoma cell lines. Our results demonstrate the replication of both transfected TTV genomes up to day 21 post transfection in all the evaluated cell lines. Quantitative amplification indicates statistically significant enhanced TTV replication in the EBV-positive cell lines, including the EBV-converted BJAB line, in comparison to the EBV-negative Burkitt's lymphoma cell line BJAB. This suggests a helper effect of EBV infections in the replication of TTV. The present study provides information on a possible interaction of EBV and TTV in the etiology and progression of multiple sclerosis.

Treatment of P3HR-1 Burkitt's lymphoma cultured cells with 4 mM n-butyrate for 48 h induced the abundant expression of a 35 000 dalton, microsomal membrane protein which was demonstrated to be the electrophoretically invariant Ii molecule. To prove this point, [35S]methionine-labelled, microsomal membrane proteins or immunoprecipitates of such detergent-solubilized proteins were subjected to electrophoresis either in SDS gels or in two-dimensional, nonequilibrium pH gradient gels. The induced p35 molecule was judged to be Ii by molecular weight and isoelectric characteristics and by immune complex associations. The abundant expression of Ii on butyrate-treated P3HR-1 cells mimicked the high level of expression of p35 or Ii on leukemic cells of a subset of patients with hairy cell leukemia. Alpha and beta chains of the HLA-D complex were absent from the P3HR-1 cell line and present in the Jijoye parent from which mutant P3HR-1 was derived. This relatively simple model can lead to the study of inducible synthesis, processing and expression of Ii, with or without HLA-D alpha and beta chains.
