Immunocytochemical evaluation of estrogen receptor on archival Papanicolaou‐stained fine‐needle aspirate smears

Abstract: The availability of limited fine-needle aspirate smears necessitates the selection of immunocytochemical (IC) methods that allow reuse of Pap-stained smears to assess the estrogen receptor (ER) status of breast carcinoma. The objective of the current study was to compare IC evaluation of ER status on FNA smears by three methods: 1) ER-ICA using H222 monoclonal antibody performed on slides fixed in formaldehyde-methanol-acetone; 2) destained Pap slides using 1D5 antibody; and 3) Pap-stained slides without desta… Show more

“…36 Even in archival Pap-stained slides, ER was detected in most cases. 37 These studies indicate that, as in tissue sections, variation in tissue fixation may not be a limiting factor; rather, antigen retrieval is essential for optimal results (Fig. C-1).…”

“…ICC on Papanicolaou-stained materials is usually possible. [1417] However, it has turned out to be quite difficult to do ICC on May Grunwaald Giemsa (MGG) or Diff-Quick ® prestained smears. Recently, an article from Choi et al .…”

“…This may be due to difference in fixation, processing, and type and length of antigen retrieval. Use of some antigen retrieval techniques optimized for histologic sections will result in cell loss from ThinPrep TM slides [24]. Furthermore, all of our postmenopausal women were on HRT and exogenous hormones use may downregulate ER expression [25].…”
