Immunobiotic lactic acid bacteria beneficially regulate immune response triggered by poly(I:C) in porcine intestinal epithelial cells

Abstract: This study analyzed the functional expression of TLR3 in various gastrointestinal tissues from adult swine and shows that TLR3 is expressed preferentially in intestinal epithelial cells (IEC), CD172a+CD11R1high and CD4+ cells from ileal Peyer's patches. We characterized the inflammatory immune response triggered by TLR3 activation in a clonal porcine intestinal epitheliocyte cell line (PIE cells) and in PIE-immune cell co-cultures, and demonstrated that these systems are valuable tools to study in vitro the im… Show more

“…The binding of dsRNA ligand poly(I·C) to TLR3, RIG-I, or MDA5 has been reported to activate the downstream signaling cascade that ends with the induction of type I interferons (IFNs) and the production of cytokines/chemokines (a T H 1 response) (10,11,14). The results of the poly(I·C)-only challenge in HT-29 cells of our study are in accordance with previously published data on the upregulation of a number of proinflammatory and antiviral genes connected to the TLR3-TRIF, NF-B, and MAPK signaling pathways (Fig.…”

“…Localized in the endosomal membrane of IECs, TLR3 triggers the activation of the innate antiviral immune response against dsRNA viruses (10,11). However, new evidence has emerged showing that two cytoplasmic PRRs of the retinoic acid-inducible gene I (RIG-I)-like receptor (RLR) family, RIG-I and melanoma differentiation-associated gene 5 (MDA5), are major sensors for dsRNA viruses, such as rotavirus, and for cell signaling of type I IFN (12,13).…”

“…Suspensions of porcine PP immunocompetent cells were prepared from the ilea of adult swine as previously described (16,31). All procedures were conducted in accordance with the Guidelines for Animal Experimentation of Tohoku University, Sendai, Japan.…”

“…These cells are permissive to porcine RVs and also respond to dsRNA and its synthetic analog poly(I:C) (49, 50). Furthermore, co-cultures of PIE cells with immune cells isolated from porcine Peyer’s patches (PPs) provide an in vitro system to study the transduction of the signal from its detection by IECs to the effect on the under laying immune cells.…”
