Viruses attach to cells via specialized attachment proteins that interact with specific cell surface receptors (1-5). An understanding of such virus-host cell interactions may provide new insights into the pathogenesis and prevention of infectious diseases (6).The mammalian reoviruses, segmented, double-stranded RNA viruses include three serotypes (7). The segmented nature of the reovirus genome has permitted a genetic approach for the identification of the role of individual viral components in pathogenesis, making the study of virus-host interactions more amenable to direct experimentation (8). These studies have revealed that each of the three outer capsid proteins (#lc, ~1, ~r3) plays critical, distinct roles in viral virulence. One of the outer capsid proteins, al, the reovirus hemagglutinin (HA), ~ has been shown to be the reovirus cell attachment protein (9-12). Studies utilizing monoclonal antibodies have revealed that the HA is divided into three distinct domains (13, 14), one of which governs the important biologic properties of receptor interaction, recognition by cytotoxic T lymphocytes (CTL) and recognition by neutralizing antibody (15,17). Reovirus mutants at this site, selected for resistance to neutralization, are altered in cell tropism (in the nervous system) (16) and in recognition by CTL (17). Therefore, monoclonal antibodies directed to this site, block a number of important biologic properties of reovirus.That such diverse biologic properties as CTL recognition and viral neutralization localize to one part of the HA suggests that binding is restricted by a unique conformation or, alternatively, by a set of epitopes on the HA. This implies that the receptor for virus on immune CTL, the natural receptor found on neurons, and the binding moiety of neutralizing monoclonal antibodies (idiotype) share a common configuration as well.To test this possibility, syngeneic monoclonal antibodies were raised against the idiotype portion of a prototypic neutralizing monocional antibody, 9BG5 (18,19). The monoclonal antibody-defined, tumorassociated antigen GA733 was purified from the SW948 human colorectal carcinoma cell line and its partial amino acid sequence was determined. By using a synthetic oligonucleotide probe, two recombinants were isolated from a total human genomic library. We prove the existence of a family of GA733 genes. One of the genomic isolates is demonstrated to be an intronless gene, which is transcribed in pancreatic carcinoma cell lines and in placenta. The GA733 proteins were observed to contain sequences homologous to a repeat unit occurring 10 times in thyroglobulin and once in the HLA-DR-associated invariant chain. A more evolutionarily distant relationship was found with the a chain of the interleukin 2 growth factor receptor.Monoclonal antibodies (mAbs) C017-1A and the related GA733 (1) have been extensively evaluated for the diagnosis and therapy of human gastrointestinal tumors. These mAbs were derived from the immunization of mice with a human colorectal adenocarcinoma cell line and a stomach adenocarcinoma cell line, respectively.

SUMMARYTo study the role of anti-idiotypic antibodies in the regulation oflhe immune response to Lolp 1 (the major allergenic component of rye grass pollen), we have recently generated a panel of three MoAbs directed against distinct epitopes of L«//71 and an anti-idiotypic MoAh directed against Ihc idiotype borne by one of the anti-Lol p I MoAbs (290A-I67). The effects of pretreatment with this aniiidiotypic MoAb in BALB/c mice before immunization wilh the antigen have been examined. The anti-idiotypic MoAb or unrelated MoAb were given weekly for 8 weeks intraperitoneally. Mice then received the antigen (2 /jg) adsorbed with alum (2 mg) at weeks 9. I! and 1.1. Scrum anti-Lf)/ p I antibodies (IgG or IgE) and specihc idiotypic responses were measured. \n\\-Lolp I IgG antibodies could be detected before immunization with Lolp I only in mice pretreated with anli-idiotypic MoAb. Immunization with Lo//? I induced an anti-L(>//)ngG response in both groups, but this response was higher in mice that received anti-idiotypic MoAb. Similar prolilcs were seen for specific IgE antibodies and idiotypic responses. Surprisingly, idiotypes borne by other anti-Ac//) I MoAbs (539A-6 and M8A-(i) had also been enhanced after pretreatment with the anti-290A-l67 MoAb. These observations suggested thai the pretreatment with this anti-idiolypic MoAb modulates not only Ihc expression of the respective idiotype. but also affects other idiotype responses.
