Immune Checkpoint Blockade Therapy: Merits and Demerits

Abstract: The up regulation of anti-tumor responses by immune-checkpoint blockade is a new approach for the treatment of advanced solid malignant tumors. There are some demerits to this therapy that require an evaluation of eligibility and efficacy, balanced against adverse effects and cost. Treatment for patients without immune checkpoint receptors or ligands remains an issue of concern. The cost of this therapy is huge and is further increased when treatment is required for adverse effects. The appropriate use of immu… Show more

“…This provides a rational for using combined BRAF/MEK inhibition instead of BRAF inhibition alone [ 11 ]. Treatment of melanoma with immune checkpoint inhibitors has a lower response rate compared to treatment with BRAF/MEK inhibitors, but the response tends to be more durable, lasting for years [ 11 13 ]. It was therefore suggested that BRAF/MEK-targeted therapy may synergize with the PD-1 pathway blockade to enhance anti-tumor immunity [ 4 , 11 , 14 , 15 ].…”

“…This can boost the body's immune response against cancer cells. Unlike radio and chemotherapy which aim to directly interfere with tumor cell growth and survival, Immunotherapy target the tumour indirectly by boosting the antitumor immune response that spontaneously arise in many patients [1] .…”
